葡萄糖及丙酮酸代谢与胚胎囊胚形成关系的研究 被引量:3

摘要 目的探讨辅助生殖技术中胚胎对培养液中葡萄糖和丙酮酸的吸收与胚胎囊胚形成的相关性。方法选择2018年9月—2019年9月在首都医科大学附属北京妇产医院行体外受精-胚胎移植(IVF-ET)的受精第3天、细胞数在5细胞及以上、分级2级以上的胚胎进行囊胚培养,将囊胚培养后形成囊胚的废弃胚胎培养液作为囊胚组,囊胚培养后未形成囊胚的废弃胚胎培养液作为非囊胚组,分别用比色法和荧光法检测2组废弃培养液中葡萄糖和丙酮酸的浓度并进行组间比较。结果囊胚组培养第3天培养液中葡萄糖浓度低于非囊胚组(P<0.05),丙酮酸浓度与非囊胚组比较差异无统计学意义(P>0.05)。囊胚组培养第5天培养液中葡萄糖、丙酮酸浓度与非囊胚组比较差异均无统计学意义(P均>0.05)。结论胚胎在培养第3天对葡萄糖高利用与囊胚形成有关,培养第3天对葡萄糖高利用提示胚胎有高发育潜能。胚胎在培养第3天及培养第5天对丙酮酸的利用与囊胚形成的关系尚不明确。 Objective It is to explore the correlation between embryonic absorption of glucose and pyruvate in culture medium and embryo blastocyst formation in assisted reproductive technology.Methods The embryos on the third day of fertilization,with a cell number of 5 cells and above,and grade 2 or above,were collected after in vitro fertilization-embryo transfer(IVF-ET)from September 2018 to September 2019 at Beijing Obstetrics and Gynecology Hospital,Capital Medical University to carry out blastocyst culture,the discarded embryo culture fluid that formed blastocysts after blastocyst culture was used as the blastocyst group,and the discarded embryo culture fluid that did not form blastocysts was used as non-blastocyst group after blastocyst culture.The concentrations of glucose and pyruvate in the waste cultures of the two groups were detected and compared between the groups.Results The concentration of glucose in culture medium in blastocyst group was lower than that of non blastocyst group after culture for 3 days,the difference between the two groups was statistically significant(P<0.05),but the difference of pyruvate concentration between blastocyst group and non blastocyst group was not statistically significant(P>0.05).There was no significant difference in the concentration of glucose and pyruvate between the blastocyst group and the non blastocyst group after culture for 5 days(P>0.05).Conclusion The high utilization of glucose in D3 is related to the formation of blastocyst.The high utilization of glucose in D3 indicates that the embryo has high developmental potential.The relationship between the utilization of pyruvate in D3 and D5 and blastocyst formation is not clear.
