七氟烷对人骨肉瘤Saos2细胞增殖、侵袭、迁移及凋亡的影响

摘要 目的探讨不同浓度七氟烷对人骨肉瘤Saos2细胞生物学行为的影响,以减少麻醉相关的肿瘤复发与转移。方法体外培养人骨肉瘤Saos2细胞,随机分为对照组、1.7%七氟烷组、3.4%七氟烷组及5.1%七氟烷组。取对数生长期Saos2细胞在不同浓度的七氟烷暴露2 h后继续培养24 h。采用cck-8法评估七氟烷对骨肉瘤Saos-2细胞增殖能力,Transwell法检测细胞侵袭能力,划痕愈合实验测定细胞的迁移能力以及流式细胞仪测定Saos2细胞凋亡。结果与对照组相比,不同浓度组的七氟烷均抑制了Saos2细胞的增殖率、细胞穿膜数、划痕愈合率,Saos2细胞的凋亡率增加,差异有统计学意义(P<0.05),随着七氟烷浓度越高,其对Saos2细胞的促进或抑制作用表现的更明显。结论七氟烷能够抑制人骨肉瘤Saos2细胞的增殖、侵袭、迁移能力,促进Saos2细胞凋亡,效应与七氟烷浓度正相关。 Objective To investigate the effects of sevoflurane at different concentrations on the proliferation and invasion of osteosarcoma cell line Saos2 in order to reduce anesthesia-related recurrence and metastasis of tumors.Me thods Human osteosarcoma Saos2 cells were cultured in vitro and randomly divided into control group,1.7%sevoflurane group,3.4%sevoflurane group and 5.1%sevoflurane group.Saos2 cells in logarithmic growth phase were exposed to sevofluraneat different concentrations for 2 hours and then cultured 24 h.CCK-8 method was used to evaluatethe cell proliferation ability.Transwell method was used to detect the change of cell invasion ability.Flow cytometry was used to determine the apoptotic rate of Saos-2 cells,and scratch healing test was used to determine the effect of sevoflurane on the migration ability of Saos2 cells.Re s ults Compared with the control group,sevoflurane in different concentration groups inhibited the proliferation rate,the number of cells membrane penetration,scratch healing rate of Saos2 cells,and the apoptotic rate of Saos2 cells increased with a statistically significant difference(P<0.05).With the higher concentration of sevoflurane,its promotion or inhibition performance on Saos2 cells is more obvious.Conclus ion Sevoflurane can inhibit the proliferation,invasion and migration of osteosarcoma saos2 cells and promote cells apoptosis,which is positively correlated with the concentration of sevoflurane.
