BMSCs膜片对静止期牙周病动物模型牙周组织IL-6表达水平的影响 被引量:2

摘要 目的探讨骨髓间充质干细胞(BMSCs)膜片局部应用对静止期牙周病动物模型牙齿移动后牙周组织IL-6表达水平的影响。方法分离、培养和鉴定BMSCs。制备BMSCs膜片。28周SD雄性大鼠随机分为对照组和实验组。对照组不做处理,实验组建立静止期牙周病动物模型,并随机分为单纯加力组(25只)和膜片+加力组(25只)。膜片+加力组局部植入BMSCs膜片,7 d后两组动物模型牙齿加力。各组于加力0,7,14,21,28 d后取右侧上颌骨,免疫荧光染色观察IL-6的表达。结果加力7,14,21,28 d,单纯加力组与膜片+加力组IL-6表达平均光密度值(AO)均较对照组明显升高(P<0.05);加力0,14,21 d,膜片+加力组AO较单纯加力组明显降低(P<0.05)。加力7 d,单纯加力组和膜片+加力组AO均达到顶峰,然后两组AO逐渐降低,但膜片+加力组降低得更快。结论BMSCs膜片的局部应用可以抑制静止期牙周病动物模型牙齿移动过程中IL-6的生成。 Objective To evaluate the effect of local application of bone mesenchymal stem cells( BMSCs) sheet on the expression of IL-6 in periodontal tissue in animal model of quiescent periodontitis. Methods BMSCs were isolated,cultured and identified. BMSCs sheet was prepared. Male SD rats aged 28 weeks were randomly divided into control group and experimental group. The rats in control group were given no treatment,and the rats in experimental group were induced to establish the animal models of quiescent periodontitis. The rats in experimental group were randomly divided into orthodontic group( n = 25) and sheet combined with orthodontic group( n = 25). The rats in sheet combined with orthodontic group were locally implanted with BMSCs sheet. After 7 d,the teeth were given orthodontic force in all animal models. The rats were sacrificed after exerting orthodontic force for 0,7,14,21,28 d to observe the expression of IL-6 by immunofluorescence staining. Results At day 0,7,14,21 and 28,the average optical densities of IL-6 expression in periodontal tissues in both orthodontic group and sheet combined with orthodontic group were significantly higher than that in control group( P < 0. 05). At day 0,14 and 21,the average optical density in sheet combined with orthodontic group was significantly lower than that in orthodontic group( P < 0. 05). The average optical density in both orthodontic group and sheet combined with orthodontic group reached the peak at day 7,and then gradually decreased,but that of the latter decreased faster. Conclusion The local application of BMSCs sheet could inhibit the production of IL-6 during the tooth movement in animal model of quiescent periodontitis.
