糖尿病大鼠骨髓干细胞和脂肪干细胞的生物学特性:对照研究 被引量:1

Biological properties of bone marrow stem cells and adipose-derived stem cells derived from T2DM rat:A comparative study

摘要 目的:探究2型糖尿病(T2DM)大鼠骨髓干细胞(BMSCs)和脂肪干细胞(ADSCs)生物学性能差异。方法:构建T2DM大鼠模型;培养ADSCs和BMSCs;检测细胞克隆能力及增殖活性;定向诱导检测成骨差异;qPCR检测成骨基因的表达;SEM、HE染色比较两种细胞膜片差异。结果:从T2DM大鼠可获得ADSCs和BMSCs;ADSCs有更强的克隆形成和增殖能力;BMSCs的成骨分化显著高于ADSCs,但ADSCs能形成较多的胶原纤维,COL-1的表达量显著高于BMSCs;ADSCs膜片可形成更多的细胞层及细胞外基质。结论:T2DM大鼠的ADSCs成骨低于BMSCs,但具有更好的增殖能力,能形成更多细胞外基质。 Objective:To study the differences in the biological properties of bone marrow stem cells(BMSCs)and adipose-derived stem cells(ADSCs)in rats with type 2 diabetes mellitus(T2MD).Methods:T2DM rat model was constructed and ADSCs and BMSCs were cultured.The ability of cell cloning and proliferation was tested.The osteogenesis differentiation was evaluated by directional differentiation induction test.The expression of osteogenic genes were analyzed by qPCR after osteogenic induction.SEM and HE staining were used to observe the morphology of the cells.Results:ADSCs and BMSCs were obtained from T2DM rats.ADSCs had higher proliferation and clone formation rate.ADSCs formed more collagen fibers while BMSCs had significantly higher osteogenetic ability.COL-1 expression in ADSCs was significantly higher than that in BMSCs.The ADSCs sheet had more cell layers and abundant extracellular matrix.Conclusion:ADSCs derived from T2DM rats have stronger proliferation and can produce more collagen fiber,while their osteogenesis is significantly lower than that of BMSCs.
