摘要
目的:探索基于钴60放射源的γ射线用于建立大鼠骨髓间充质干细胞实验模型的理想加载条件。方法:分别应用γ射线和X射线对大鼠骨髓间充质干细胞(BMSCs)进行梯度剂量(2~8 Gy)照射,照射后检测各剂量下细胞增殖能力和凋亡程度;并检测照射后各剂量下活细胞比例和NOX4合成量,进行组间比较。结果:4 Gy以上剂量γ射线可造成细胞形态、增殖能力下降、凋亡增加;4 Gy剂量亚组活细胞明显高于6、8 Gy剂量亚组;相同剂量的γ和X射线在抑制细胞增殖、促细胞凋亡、促NOX4合成等方面无显著性差异。结论:利用钴60放射源的γ射线在4 Gy剂量、2.538 Gy/min剂量率照射条件下可建立大鼠骨髓间充质干细胞实验模型,与同等剂量的X射线具有较好的等效关系。
Objective:To explore the ideal loading conditions for the establishment of experimental model of rat bone marrow mesenchymal stem cells(BMSCs)based onγ-ray of Co 60.Methods:BMSCs of rats were irradiated byγ-ray and X-ray at 2-8 Gy respectively.After irradiation,the proliferation and apoptosis of the cells were detected by CCK-8 assay and TUNEL staining respectively,the ratio of viable cells and the amount of NOX4 synthesis after irradiation were measured by immunofluorescence technique and LCSM respectively.Results:When the dose ofγ-ray was more than 4 Gy,the cell morphology,proliferation and apoptosis were significantly inhibited;the living cell stock of 4 Gy group was significantly higher than that of 6 Gy and 8 Gy group;there was no significant difference betweenγ-ray and X-ray at the same dosage in inhibiting cell proliferation,promoting cell apoptosis and promoting NOX4 synthesis.Conclusion:The experimental model of BMSCs can be established under the irradiation conditions of 4 Gy dose and 2.538 Gy/min dose rate usingγ-ray of Co 60 radiation source,which has a good equivalent relationship with the same dose of X-ray.
