Study on Molecular Mechanism of Nanxu Yimu Tablet in Treatment of Rheumatoid Arthritis Based on Network Pharmacology

摘要 目的基于网络药理学研究探讨南续益母片治疗类风湿关节炎的分子机制。方法使用TCMSP和TCMID数据平台筛选南续益母片的有效成分及靶点,通过TTD和DRUGBANK及DisGeNET数据库检索类风湿关节炎的疾病靶点,使用Bioinformatics&Evolutionary Genomics软件筛选关键靶点,运用ClueGO插件和EHBIO生物信息软件进行KEGG通路富集分析,使用STRING软件平台和Cytoscape软件构建成分-靶点-通路和蛋白互作网络。结果最终筛选出南续益母片有效成分13个,预测治疗靶点228个,作用于类风湿关节炎的重要靶点51个,通路富集分析显示南续益母片主要作用在与类风湿关节炎相关的炎症、免疫及细胞凋亡等信号通路。结论南续益母片具有多成分、多靶点的作用特点,通过影响炎症、免疫、细胞凋亡等方面发挥治疗类风湿关节炎的作用。 Objective To investigate the molecular mechanism of Nanxu Yimu Tablet in the treatment of rheumatoid arthritis based on network pharmacology. Methods The TCMSP and TCMID data platforms were used to screen the active components and targets of Nanxu Yimu Tablet. The targets of rheumatoid arthritis were searched by TTD and DRUGBANK and DisGeNET databases, and key targets were screened using Bioinformatics & Evolutionary Genomics software. The ClueGO plug-in and EHBIO bioinformatics software performed KEGG pathway enrichment analysis, using the STRING software platform and Cytoscape software to construct a component-target-pathway and protein interaction network. Results Finally, 13 active components of Nanxu Yimu Tablet were screened, and 228 therapeutic targets were predicted. A total of 51 important targets for rheumatoid arthritis were analyzed. Pathway enrichment analysis showed that Nanxu Yimu Tablet acted on rheumatoid joints. Pathway enrichment analysis showed that Nanxu Yimu Tablet mainly acted on signaling pathways related to rheumatoid arthritis, such as inflammation, immunity and apoptosis. Conclusion Nanxu Yimu Tablet has the multi-component and multi-target characteristics, and plays a role in the treatment of rheumatoid arthritis by affecting inflammation, immunity and apoptosis.
