基于血清代谢组学的天麻炮制前后对小鼠生物学效应影响 被引量:4

Influence of Gastrodia Elata on Biological Effects before and after Processing Based on Serum Metabolomics in Mice

摘要 目的:本研究以中药整体观为指导,运用血清代谢组学的方法,从整体代谢角度分析天麻生药与饮片干预小鼠后可能存在的血清内源性代谢物的变化,筛选并鉴别出差异代谢物和代谢途径,从而评价和探讨天麻炮制前后对于机体代谢的影响。方法:通过不同时段(1,3,5 d)灌胃小鼠,利用LC-MS技术检测不同时段的小鼠血液代谢图谱,采用主成分分析(PCA)和偏最小二乘判别分析(PLS-DA)法,变量重要性投影(VIP)和t检验筛选潜在差异代谢标志物,从整体代谢角度分析天麻炮制前后生物学效应的差异。结果:通过对小鼠体内代谢物质以及代谢通路的分析,确定15个差异代谢物。在血清代谢的层面,炮制后天麻与生药有显著的生物学效应差别。结论:天麻生药经过炮制以后,会引起氨基酸代谢、肠道菌群代谢等多条代谢通路在不同层面协同发挥作用,具有多靶点性,产生与天麻生药不同的功效,为探讨天麻炮制增效机制提供理论和数据支撑。 Objective This study was guided by the holistic view of traditional Chinese medicine and used the serum metabolomics method to analyze the changes of serum endogenous metabolites that may exist after the intervention of Gastrodia elata and decoction tablets in mice,thereby screening and identifying the differential metabolic pathways,so as to evaluate and explore the influence of Gastrodia Elata on body metabolism.Methods Gavage mice through different periods(1,3,5 days)and use LC-MS technology to detect the blood metabolism profiles of mice in different times.Principal component analysis(PCA),partial least squares discriminant analysis(PLS-DA),variable importance projection(VIP),and t-test were used to screen potential differential metabolic markers.The biological effects of Gastrodia elata before and after processing were analyzed from the perspective of overall metabolism.Results 15 differential metabolic markers were determined by the analysis of metabolites and metabolic pathways in mice.At the level of serum metabolism,there were significant differences in biological effects between processed Gastrodia elata and crude drugs.Conclusion After the Gastrodia crude drug is processed,it will work synergistically at different levels and multiple metabolic pathways with multiple targets such as amino acid metabolism and intestinal flora metabolism,which is different from crude Gastrodia elata.This study provides theoretical and data support for exploring the mechanism of processing efficiency of Gastrodia elata.
