星点设计-效应面法优化黄芩苷两种提取工艺并比较 被引量:4

Optimization and Comparison of Two Extraction Processes of Baicalin by Central Composite Design-Response Surface Methodology

摘要 目的:对比回流提取法和超声提取法提取黄芩中黄芩苷的效率,并对提取工艺进行优化。方法:以黄芩苷得率为指标,在单因素试验的基础上,采用星点设计-效应面法考察乙醇浓度、液料比、提取时间对黄芩苷得率的影响。结果:回流提取法最佳提取工艺为:乙醇浓度77%、液料比10∶1、提取时间2 h、提取2次,黄芩苷得率12.55%。超声提取法最佳的提取工艺为:乙醇浓度77%、液料比22∶1、提取时间52 min、提取1次,黄芩苷得率8.32%。回流提取法的黄芩苷得率远大于超声提取法,因此回流提取法优于超声提取法。结论:回流提取法简单易操作、成本低,采用星点设计-效应面法优化的黄芩苷提取工艺稳定可行、方法简便、预测性好,可为生产工艺提供参考。 Objective The extraction efficiency of baicalin from Scutellaria baicalensis Georgi by reflux extraction and ultrasonic extraction was compared,and the extraction process was optimized.Methods On the basis of single factor experiment and taking baicalin yield as the index,the influence of ethanol concentration,ratio of liquid-material and extraction time on the yield of baicalin was studied using response surface methodology.Results The optimal extraction process of reflux extraction method was:77%ethanol concentration,10∶1 ratio of liquid-material,2 hours of extraction time,2 times of extraction.The yield of baicalin was 12.55%.The optimal extraction process of ultrasonic extraction methodis:77%ethanol concentration,22∶1 ratio of liquid-material,52 minutes of extraction time,one extraction.The yield of baicalin was 8.32%.The yield of baicalin by reflux extraction is much higher than that by ultrasonic extraction,so reflux extraction is better than ultrasonic extraction.Conclusion The reflux extraction method is simple and easy to operate with low cost.The extraction process of baicalin optimized by the central composite design-response surface methodology is stable,feasible,simple and predictable.It can provide reference for production process.
