IL-38对胶原诱导性关节炎大鼠Th17/Treg失衡的调节作用

Regulation of IL-38 on Th17/Treg imbalance in collagen-induced arthritis rats

目的探讨IL-38对胶原诱导性关节炎Th17/Treg失衡的调节作用。方法SPF级SD大鼠30只,随机分为3组,即正常对照组(CON)、胶原诱导性关节炎大鼠(collagen-induced arthritis rats,CIA)、CIA+IL-38(5ng/g)组;CIA组、IL-38组采用大鼠尾根部皮下注射牛II型胶原乳剂建立CIA大鼠模型;IL-38组于初次免疫后第24天起每天注射IL-38(20-152)5 ng/g,连续给药10d;观察大鼠后足及关节情况,HE染色观察大鼠滑膜组织病理变化,流式检测Th17/Treg细胞变化,ELISA、qRT-PCR检测Th17/Treg相关基因的表达情况。结果IL-38明显改善胶原诱导性关节炎大鼠的症状、滑膜组织病理情况、调节Th17/Treg细胞的表达水平以及Th17/Treg相关基因的表达水平。结论IL-38通过调节Th17/Treg失衡对胶原诱导性关节炎大鼠具有保护作用。

Objective To investigate the regulation of IL-38 on Th17/Treg imbalance in collagen-induced arthritis(CIA)and its mechanism.Methods Thirty SPF-grade SD rats were randomly divided into 3 groups:normal control group(CON),collagen-induced arthritis rats(CIA),CIA+IL-38(5 ng/g)group.CIA group and IL-38 group were established by subcutaneous injection of bovine type II collagen emulsion in rat tail root to establish CIA rat model;IL-38 group was daily from 24 th day after initial immunization.Recombinant murine IL-38(20-152)5 ng/g was injected for 10 days.The hind paws and joints of rats were observed.The pathological changes of synovial tissue were observed by HE staining.The changes of Th17/Treg cells were detected by flow cytometry.The expression of Th17/Treg related genes was detected by ELISA and qRT-PCR.The expression of Cause.Results IL-38 could improve the symptoms,synovial histopathology,regulate Th17/Treg cell expression and Th17/Treg related gene expression in collagen-induced arthritis rats.Conclusion IL-38 can protect collagen-induced arthritis rats by regulating the imbalance of Th17/Treg.
