摘要
目的检测前列腺组织中NUCB2和Bcl-2蛋白的表达,探讨二者在前列腺癌中表达的意义和相关性。方法收集选取本院2010年1月至2019年1月活检或手术切除的患者前列腺组织标本及临床资料,应用免疫组化SP法检测在良性前列腺增生(benign prostatic hyperplasia,BPH)、前列腺上皮内瘤变(prostatic intraepithelial neoplasia,PIN)及前列腺癌(prostate cancer,PCa)组织中NUCB2与Bcl-2的表达,并分析二者在PCa组织中的表达有无相关性。结果 NUCB2在PCa组织中的表达显著高于BPH和PIN组织,其表达与年龄无关,与Gleason分级、术前血清PSA有关;生存期小于5年病例的NUCB2表达阳性率高于生存期大于、等于5年病例(P<0.05)。Bcl-2在PCa组织中的表达显著高于BPH和PIN组织,其表达与年龄无关,与Gleason分级及血清PSA均有关。生存期小于5年病例的bcl-2表达阳性率高于生存期大于、等于5年病例,但无统计学差异(P>0.05)。在前列腺癌组织中NUCB2与Bcl-2的表达呈正相关(P=0.012)。结论 NUCB2与Bcl-2两者间可能存在一种相关性,联合检测二者有助于前列腺癌的进一步研究。NUCB2的检测可能有助于评估前列腺癌的预后。
Objective To detect the expressions of NUCB2 and Bcl-2 protein in prostate cancer tissues, and explore their significance and correlation in prostate cancer. Methods Prostate tissues of the patients with BPH,PIN or PCa, who underwent the biopsy or surgical resection in our hospital from January 2010 to January 2019, were collected. Clinical data of the patients were retrospectively analyzed. The expressions of NUCB2 and Bcl-2 in BPH, PIN and PCa tissues were detected by immunohistochemistry, and their correlation and significance in PCa tissue were further clarified. Results The expression of NUCB2 in PCa tissues was significantly higher than that in BPH and PIN tissues, and its expression level was related to Gleason grade and preoperative serum PSA, but not related to age. The positive rate of NUCB2 expression in 5-year survival cases was higher than ≥5 years survival cases(P<0.05).The expression of Bcl-2 in PCa tissues was significantly higher than that in BPH and PIN tissues, and its expression was not related to age, and was related to Gleason grade and serum PSA. The positive rate of bcl-2 expression in 5-year survival cases was higher than that in ≥5 years survival, but there was no statistical difference(P>0.05).There was a positive correlation between NUCB2 and Bcl-2 expression in prostate cancer tissues(P= 0.012). Conclusions There may be a positive correlation between NUCB2 and Bcl-2. NUCB2 expression may be helpful in assessing the prognosis of prostate cancer. Co-expression analysis of NUCB2 and Bcl-2 may contribute to further research of prostate cancer.
