β-胡萝卜素对小鼠骨髓源树突状细胞成熟的影响

摘要 为探讨β-胡萝卜素对小鼠骨髓源树突状细胞(BMDCs)活化和成熟的影响,从小鼠骨髓中分离获得前体细胞, GM-CSF和IL-4诱导使其分化成未成熟的BMDCs,并用不同浓度的β-胡萝卜素进行作用,检测BMDCs的表型标志、吞噬能力以及细胞因子的分泌水平。β-胡萝卜素作用后BMDCs进一步与异种CD4^+T细胞混合培养,检测T细胞的增殖情况。结果表明:与未处理组相比,经β-胡萝卜素作用24 h后, BMDCs表面分子CD40、MHCII、CD80和CD86的表达量显著上调, BMDCs吞噬FITC-Dextran的能力显著下降,细胞因子IL-12p70和IL-10的分泌水平显著上升, BMDCs刺激CD4^+T细胞的增殖能力显著增强。这说明β-胡萝卜素可促进DCs的活化和成熟,可作为潜在的疫苗佐剂。 To investigate the effects of β-Carotene on the activation and maturation of mouse bone marrow-derived dendritic cells(BMDCs), immature BMDCs were generated from GM-CSF and IL-4-induced progenitor cells. Subsequently, different concentrations of β-Carotene were directly interacted with BMDCs. Then, the expression of phenotype markers, the ability of endocytosis, and the secretion levels of cytokines were detected. For mixed lymphocyte reaction, the collected BMDCs were cocultured with CD4^+ T cells, and the T cell proliferation was detected. In Comparison with medium, β-Carotene remarkably up-regulated the expression of CD40, MHCII, CD80, and CD86. Similarly, β-Carotene significantly increased the release of cytokines(IL-12 p70 and IL-10). In addition, the ability of uptake FITC-Dextran was lower after 24 h treatment with β-Carotene(P<0.05). Finally, the BMDCs from β-Carotene group strongly enhanced the proliferation of allogeneic T cells compared to that from medium group(P<0.05). These results suggested that β-Carotene stimulated the activation and maturation of BMDCs, which implied that β-Carotene can be used as a potential vaccine adjuvant.
