高通量测序研究德昂酸茶对高脂饮食小鼠肠道微生物的影响 被引量:12

摘要 为探究德昂酸茶对高脂饮食小鼠的抗肥胖作用,按体重分别以德昂酸茶1.0 (低)、2.0 (中)、3.0 (高) g·kg^-1剂量对高脂饮食小鼠干预12周后,采集粪便,利用16S rDNA测序对肠道微生物菌群多样性进行分析。结果表明:德昂酸茶干预后,可显著提高高脂饮食小鼠肠道菌群的物种丰度和多样性;高脂饮食小鼠肠道优势菌群厚壁菌门丰度下降,拟杆菌门丰度提高;双歧杆菌属在各组中比例增加,空白对照组和酸茶低剂量组比例相近;另枝菌属在高脂对照组中含量较高(2.24%),但在酸茶组中,随着灌胃剂量的升高呈递减趋势。高脂对照组与其余各组肠道菌群的主成分区分明显,酸茶组菌群组成结构发生偏移。这一研究提示德昂酸茶可维持高脂饮食小鼠肠道微生态的平衡,降低厚壁菌门与拟杆菌门的比值,对减肥具有积极的意义。 In order to explore the anti-obesity effects of De’ang pickled tea on high-fat diet mice, high-fat diet mice were intervented with De’ang pickled tea in low dose(1.0 g·kg^-1), medium dose(2.0 g·kg^-1) and high dose(3.0 g·kg^-1) for 12 weeks. Then, the feces of mice were collected and the fecal microbial diversity was analyzed by 16 S rDNA sequencing. The results showd that the species abundance and diversity of intestinal flora of high-fat diet mice could be significantly increased after the intervention of De’ang pickled tea(P<0.05). The abundance of dominant flora Firmicutes in the intestinal flora of high-fat mice decreased, while Bacteroidetes increased correspondingly. The proportion of Bifidobacterium increased in each group, and the blank control group was similar to the Pickled tea low-dose group. In addition, the content of Alitise was higher in the high-fat control group(2.24%), and the Pickled tea group showed a decreasing trend with the increase of gavage dose. The principal components of the high-fat control group were clearly distinguished from the rest of the groups, and the composition structure of the bacteria of the Pickled-tea group was somewhat deviated. De’ang pickled tea can maintain the intestinal micro-ecological balance, reduce the ratio of Firmicutes to Bacteroides and has positive significance for weight loss.
