摘要
以脱乙酰度90%的壳聚糖为原料,采用两步合成方法,得到N,N,N-三甲基壳聚糖(TMC),将TMC与氯乙酸反应,得到端羧基季铵化壳聚糖(CTMC),利用傅里叶变换红外光谱仪和核磁共振氢谱仪对产物进行了表征并研究其对大肠杆菌和金黄色葡萄球菌的抗菌性能。结果表明:通过两步法成功合成了CTMC,实现了对壳聚糖分子结构的改性;CTMC的溶解性得到明显提高,其可以溶解于任意pH的水溶液中,且在中性溶液中表现出一定的抗菌性。CTMC的抗菌机理可能主要是带正电的壳聚糖分子链与带负电的细菌细胞壁存在静电作用,改变了细胞壁的通透性,影响细胞内外物质传递从而杀死细菌。
N,N,N-trimethyl chitosan(TMC)was successfully synthesized making use of two steps method,and carboxymethyl-quaternized chitosan(CTMC)was further obtained by TMC reacted with monobromoacetic acid.The products were characterized and their performance against E.coli and S.aureus were studied.The results indicated that CTMC was successfully synthesized and the molecular structure of CS was modified,CTMC could dissolve in aqueous solution with any pH value and exhibited antibacterial activity in neutral solution.In addition,the antibacterial mechanism may be that the positive charged chitosan can adsorb to the negative charged surface of bacterial,which changed the permeability of the cell wall,influenced the cytoplasm flow of the cell,and finally kill bacteria.
