灯台叶碱调节放射性肺炎细胞及大鼠血清细胞因子水平的实验研究 被引量:2

Study on the Regulation of Cytokine Levels in Radiation Pneumonitis Cells and Rat Serum by Alkaloids from Leaves of Folium Alstoniae Scholaris

摘要 目的探讨灯台叶碱对放射性肺炎细胞及大鼠血清细胞因子水平的调节作用,为灯台叶制剂的临床应用及放射性肺炎的临床治疗提供新思路。方法通过建立放射性肺炎细胞及大鼠动物模型,检测不同浓度灯台叶碱对细胞上清液及大鼠血清中细胞因子TNF-α、IL-2、IL-4、IL-6、IL-10水平的影响。结果放射治疗后灯台叶碱干预的BEAS-2B细胞上清中,炎症介质IL-2、IL-4、IL-10和TNF-α水平降低(P<0.05),IL-6在灯台叶碱处理前后未见明显变化。放射治疗后灯台叶碱干预的大鼠血清中IL-2、TNF-α等促炎因子下降(P<0.05),但是对血清炎症介质的抑制程度无明显剂量依赖性。灯台叶碱治疗组可升高血清抗炎因子IL-4的水平(P<0.05),而其升高作用无显著的剂量依赖性。结论灯台叶碱可能通过降低血清促炎介质的水平和升高抗炎介质的血清水平抑制放射性肺炎的炎性反应,有望成为防治放射性肺炎的有效药物。 Objective To investigate the regulatory effect of alkaloids from the leaves of Folium Alstoniae Scholaris on the cytokine levels in the radiation pneumonitis cells and rat serum,so as to provide evidence for the clinical application of alstonia-leaf alkaloids preparations and to supply new way for the treatment of radiation pneumonitis.Methods The cell models and rat model of radiation pneumonia were established for detecting the effects of different concentrations of alstonia-leaf alkaloids on the levels of cytokines of TNF-α,IL-2,IL-4,IL-6 and IL-10 in cell supernatant and rat serum.Results The levels of IL-2,IL-4,IL-6,IL-10 and TNF-αwere decreased in the supernatant of BEAS-2B cells treated with alstonia-leaf alkaloids after radiotherapy(P<0.05),but the level of IL-6 showed no significant changes after intervention with the alstonia-leaf alkaliods.The levels of proinflammatory cytokines of IL-2 and TNF-αwere decreased in the serum of rats treated with alstonia-leaf alkaloids after radiotherapy(P<0.05),but the dose-dependent effect on the serum cytokines was not obvious.In the intervention group with alstonia-leaf alkaloids after radiotherapy,the serum level of anti-inflammatory factor(IL-4)was increased with time(P<0.05),but the dose-dependent effect on the serum IL-4 was not obvious.Conclusion The alstonia-leaf alkaloids may achieve the therapeutic effect for radiation pneumonitis by reducing the serum levels of pro-inflammatory mediators and increasing serum levels of anti-inflammatory mediators,andthe alkaloids are expected to become an effective drug for radiation pneumonitis.
