基于网络药理学探讨金荞麦治疗慢性咽炎的作用机制 被引量:3

摘要 目的:通过网络药理学方法探究金荞麦治疗慢性咽炎的有效成分、关键靶点、相关通路信息。方法:利用中药系统药理学数据库与分析平台TCMSP数据库检索,找出药物金荞麦的主要成分及作用靶点;利用人类基因组数据注释系统(Gene Cards)和联机孟德尔人类遗传数据库(OMIM)找出与疾病相关的作用靶点,将金荞麦的主要成分的作用靶点与慢性咽炎的相关靶点取交集,将交集靶点导入STRING数据库得到PPI网络,对该网络进行拓扑分析找出关键靶点,且通过Cytoscape3.7.0软件构建"药物-主要成分-疾病-作用靶点"网络图;把关键靶点导入DAVID数据库,找出金荞麦治疗慢性咽炎关键靶点参与的生物学过程、分子功能、细胞组分和KEGG通路。结果:通过TCMSP数据库的口服利用度(OB)和类药性(DL)条件筛选出15个金荞麦主要化合物,相应作用靶点100个;与慢性咽炎发病机制有关的作用靶点40个,经拓扑分析得到14个关键靶点;GO分析富集得到生物过程109个,分子功能30个,细胞组分9个,47条KEGG信号通路。结论:采用网络药理学的方法初步判断金荞麦治疗慢性咽炎是多成分-多靶点-多通路实现的,可进一步揭示金荞麦的药用价值。 Objective To explore the information of active ingredients,key targets and related pathways of golden buckwheat in the treatment of chronic pharyngitis by network pharmacology.Methods The Chinese medicine system pharmacology database and analysis platform TCMSP database were used to find out the main components and target of the drug gold buckwheat;Using the Human Genome Data Annotation System(Gene Cards)and the Online Mendelian Human Genetic Database(OMIM)to find disease-related action targets,intersect the action targets of the main components of golden buckwheat with related targets of chronic pharyngitis,The intersection target is imported into the STRING database to obtain a PPI net^vork,and the network is subjected to topology analysis to find key targets,and a“drug-main component-disease-acting target”network diagram is constructed by Cytoscape 3.7.0 software;The key targets were imported into the DAVID database,and the biological processes,molecular functions,cellular components,and KEGGpathways involved in the key targets of golden buckwheat tor chronic pharyngitis were found.Results According to the oral availability(OB)and drug-like(DL)conditions of the TCMSP database,15 major buckwheat compounds were screened out,corresponding to 100 targets;40 targets were related to the pathogenesis of chronic pharyngitis.Fourteen key targets were obtained by analysis;109 biological processes,30 molecular functions,9 cellular components,and 47 KEGG signaling pathways were obtained by GO analysis and enrichment.Conclusion Using network pharmacology method to preliminarily judge that the treatment of chronic pharyngi-tis by golden buckwheat is realized by multi-component-multi-target-multi-path,which can further reveal the medicinal value of golden buckwheat.
