百香果叶片植株再生快繁技术 被引量:8

Rapid Propagation Technology of Plant Regeneration from Leaves of Passiflora edulis Sims

摘要 以百香果种子无菌实生苗的子叶、真叶及田间茎尖嫩叶为外植体,以MS为基本培养基,经愈伤组织诱导、丛生芽分化、腋芽增殖及生根培养4个阶段进行比较试验,以探究适宜的外植体种类及4个培养阶段中最佳的激素种类、浓度和配比,为百香果组培快繁技术提供参考依据。结果表明,3种外植体中,子叶最容易诱导产生愈伤组织,其诱导率为100%;最佳子叶愈伤组织诱导培养基为:MS+6-BA 1.5 mg/L+NAA 0.2 mg/L;最佳不定芽分化培养基为:MS+6-BA 1.0 mg/L+NAA 0.2 mg/L;最佳腋芽增殖培养基为:MS+6-BA 1.0 mg/L+IBA 0.2 mg/L;最佳生根培养基为:MS+NAA 0.3 mg/L。 The cotyledon,true leaf and shoot tip tender leaf of the aseptic seedling of passion fruit seed were used as explants and MS as the basic medium,a comparative experiment was conducted in four stages callus induction,cluster bud differentiation,axillary bud proliferation and root culture,to explore the suitable explant species and the optimal hormone types,concentrations and proportions in the four culture stages,and provide a reference for tissue culture and rapid propagation of Passiflora edulis Sims.The results showed that among the three explants,cotyledons were the most prone to induce callus formation,with the induction rate of 100%.The best medium for callus induction was MS+6-BA 1.5 mg/L+NAA 0.2 mg/L,the best medium for cluster bud differentiation was MS+6-BA 1.0 mg/L+NAA 0.2 mg/L,the optimum medium for adventitious bud differentiation was MS+6-BA 1.0 mg/L+IBA 0.2 mg/L,the optimum medium for rooting was MS+NAA 0.3 mg/L.
