摘要
目的:建立适用于心肌成纤维细胞体外分离、培养及鉴定的技术。方法:细胞差数贴壁法培养SD大鼠乳鼠心肌成纤维细胞,免疫荧光法鉴定。结果:心肌成纤维细胞排列紧密,交叉重叠生长,无自发性搏动,到80%~90%时可传代。结论:体外培养心肌成纤维细胞以及免疫荧光鉴定可行、有效,为临床研究心脏疾病提供理想的实验模型。
Objective:To establish techniques which suitable for isolation,culture and identification of cardiac fibroblasts in vitro.Methods:Suckling rats of SD rats'myocardial fibroblasts were cultured by cell difference attachment method and identified by immunofluorescence.Results:Myocardial fibroblasts were closely arranged,with cross-overlapping growth,no spontaneous pulsation,to 80%~90%can be subcultured.Conclusion:In vitro culture of myocardial fibroblasts and immunofluorescence identification are feasible and effective to provide an ideal experimental model for clinical study of heart disease.
