复合应激因素对应激性高血压大鼠肾脏的损伤作用及意义 被引量:1

The effect and significance of compound stress factors on kidney injury of SIH rats

摘要 目的在复合应激因素作用下,成功建立应激性高血压(stress-induced hypertention,SIH)大鼠模型,探讨SIH大鼠模型肾脏损伤的机制在SIH发生、发展过程中的作用,为应激性高血压肾脏损伤的防治提供实验依据。方法建立应激性高血压大鼠模型,记录左颈总动脉血压值,放射免疫法检测血浆皮质醇及内皮素含量的变化;光镜下观察肾脏的形态学变化以及Masson染色结果;采用速率散射比浊法测定尿微量蛋白水平;利用免疫散射比浊法检测尿蛋白水平。结果(1)成功建立应激性高血压大鼠模型,与正常对照组相比,SIH模型组大鼠收缩压和舒张压均明显升高(P<0.05),应激相关激素皮质醇及内皮素含量明显升高(P<0.05)。(2)光镜结果显示,SIH模型组大鼠肾脏组织肾小球固缩,肾小囊间隙增宽;Masson结果显示,SIH模型组大鼠肾间质,肾小管及肾小球可见大量染成蓝色的胶原纤维;(3)24 h尿蛋白与尿微量蛋白水平显著升高(P<0.05)。结论本研究成功建立了应激性高血压大鼠模型,应激性高血压大鼠出现肾脏损伤的病理改变。 Objective To establish stress-induced hypertension(SIH)rat model successfully by effects of combined stress factors,and to explore the role of renal injury mechanism in the development of SIH,so as to provide experimental basis for prevention and treatment on renal injury of SIH.Methods The model of stress hypertensive rats was established.The blood pressure of left common carotid artery was recorded.The changes of plasma cortisol and endothelin were detected by radioimmunoassay.The morphological changes of kidney were observed by HE and Masson staining.The level of urine microprotein was measured by rate scattering turbidimetry and the level of urine protein was measured by immune scattering turbidimetry.Results(1)Compared with the normal group,the systolic and diastolic blood pressure in the SIH group increased significantly(P<0.05),and the contents of cortisol and endothelin increased significantly(P<0.05),(2)In the SIH group,the HE staining results showed that the glomerulus was pyknosis and the renal vesicle space was widened,Masson staining results showed that a large number of blue stained collagen fibers could be seen in the renal interstitium,renal tubules and glomeruli,(3)The level of 24-hour urinary microprotein and urinary protein increased significantly(P<0.05).Conclusion The model of stress-induced hypertension was established successfully in this study;The pathological changes of renal injury were found in the stress-induced hypertension rats.
