miR-101对宫颈癌HeLa细胞裸鼠皮下移植瘤生长及肺转移的影响 被引量:1

摘要 目的建立宫颈癌HeLa细胞裸鼠皮下移植瘤及肺转移瘤模型,观察miR-101对HeLa细胞裸鼠皮下移植瘤及肺转移瘤形成的作用。方法90只清洁雌性BALB/c裸鼠,周龄(4~6)周。通过构建miR-101慢病毒载体转染宫颈癌HeLa细胞。其中,45只裸鼠建立皮下移植瘤模型,分为空白组、对照组、实验组,每组15只。空白组注射未处理的HeLa细胞;对照组注射空载慢病毒转染的HeLa细胞;实验组注射携带miR-101的慢病毒载体转染的HeLa细胞。另45只裸鼠建立肺转移瘤模型,分组同皮下移植瘤模型。观察肿瘤体积、成瘤裸鼠体质量及一般情况,并制作病理组织学切片,观察肿瘤及重要器官的组织学特点。结果皮下移植瘤实验结果显示,实验组肿瘤体积低于空白组及对照组,差异具有统计学意义(P<0.05)。组织切片图可见移植瘤呈乳头状,与周围组织界限清晰;部分实验组移植瘤可见坏死区。其病理组织特点为肿瘤细胞符合HeLa细胞的形态特点。空白组及对照组可见肿瘤细胞有显著异型性,部分实验组可见呈颗粒状红染的坏死组织。肺转移瘤实验结果显示,实验组裸鼠体质量高于空白组及对照组,差异具有统计学意义(P<0.05)。其病理组织特点为实验组肺转移瘤呈小灶状分布,可见坏死细胞。结论高表达miR-101可抑制宫颈癌HeLa细胞裸鼠皮下移植瘤的生长。 Objective To observe effects of miR-101 on the growth of subcutaneous xenograft and lung metastasis of HeLa cervical carcinoma cells in nude mice through building the corresponding model.Methods A total of 90 female(4-6)week-old clean BALB/c nude mice were used and lentivirus transduced HeLa cervical cancer cells carrying miR-101 were constructed.The model of subcutaneous transplantation tumor was established in 45 nude mice,which were divided into three groups with 15 per each:blank group(untreated HeLa cells),control group(no-load lentivirus transduced HeLa cells),and experimental group(lentivirus transduced HeLa cells carrying miR-101).Another 45 nude mice were developed the model of lung metastasis,grouping with the same to the subcutaneous transplantation tumor model's.The tumor volume,quality and general situation of the tumor-bearing nude mice were observed,and histological features of the tumors and important organs were analyzed with HE staining.Results Subcutaneous xenograft experiment showed that the tumor volume in the experimental group was lower than that in the blank group and the control group,and the difference was statistically significant(P<0.05).The gross appears to have papillary transplanted tumor and clear boundaries with the surrounding tissues;necrotic areas were also observed in the experimental group,whose pathological features were that tumor cells conform to the morphological characteristics of HeLa cells.Distinctive atypia from the tumor cells was shown in the blank group and the control group,and the necrotic tissue with granular red staining in the experimental group.Lung metastasis experiment showed that the weight of the nude mice in the experimental group was higher than that in the blank group and the control group,and the difference was statistically significant(P<0.05);the pathological features were small focal distribution of lung metastases and necrotic cells were visible.Conclusion High expression of miR-101 could inhibit the growth of subcutaneous xenografts of cervical cancer HeLa cells in nude mice.
