雪莲注射液对切口痛大鼠痛觉及致痛物质水平的影响

Effects of Xuelian injection on pain sensation and algogenic substances in rats undergoing the incision pain

摘要 目的研究腹腔内注射雪莲注射液对足底切口痛模型大鼠的镇痛作用,及对致痛物质P物质(SP)、神经激肽1-受体(NK-1R)水平的影响。方法成年雄性SD大鼠96只随机分成3组:曲马多注射液组(T组)、雪莲注射液组(S组)、模型组(C组),每组32只,制作足底切口痛大鼠模型。手术完成后行腹腔给药,T组:曲马多10 mg/kg,S组:雪莲注射液0.72 mL/kg,C组:等容量生理盐水。检测手术前1 d(T0)及手术后1 h(T1)、2 h(T2)、4 h(T3)、8 h(T4)不同时点大鼠机械痛阈的变化,采用酶联免疫吸附试验(ELISA)法检测大鼠血清P物质(SP)及神经激肽-1受体(NK-1R)的变化,采用Western blot法检测大鼠脊髓SP及NK-1R蛋白表达。结果手术前各组之间机械痛阈无差异,手术后3组大鼠机械痛阈均较手术前降低(P<0.05);其中,S组和T组相比无统计学差异(P>0.05);C组较S组和T组痛阈降低(P<0.05);血清SP、NK-1R含量与脊髓SP、NK-1R表达在S组和T组相比差异无统计学意义(P>0.05),两组均较C组降低(P<0.05)。结论雪莲注射液与曲马多注射液均对切口痛模型大鼠镇痛有效,镇痛效果无统计学差异,雪莲注射液可替代曲马多作为中重度疼痛的镇痛选择。 Objective To study the analgesic effect of Xuelian injection in rats undergoing plantar incision pain and the levels of substance P(SP),neurokinin 1-receptor(NK-1R)in serum and spinal cord.Methods A total of 96 adult male SD rats were received operation to establish the model of plantar incision pain,then randomly divided into 3 groups(n=32 each):tramadol injection group(Group T),Xuelian injection group(Group S)and control group(Group C).Rats in the groups were received tramadol injection 10 mL/kg,Xuelian injection 0.72 mL/kg,and equal volume of normal saline,respectively.Paw withdrawal mechanical threshold(PWMT)in rats were detected and recorded before the operation and after 1 h(T1),2 h(T2),4 h(T3),8 h(T4)of the operation.Then the blood and spinal cord samples were collected,and the mRNA and protein levels of SP and NK-1R were measured by ELISA and Western blot respectively.Results There were no differences in PWMT between the groups before surgery.The PWMT decreased in each group after surgery(P<0.05),and there was no statistical difference between the Group S and Group T(P>0.05).Compared with the Group C,the PWMT were significantly increased in the Group S and Group T(P<0.05).Compared with the Group C,the levels of SP and NK-1R in the serum and protein expression in the spinal cord were decreased and down-regulated in the Group S and Group T(P<0.05).There was no significant difference between the Group S and Group T(P>0.05).Conclusion Xuelian injection and tramadol injection showed equal analgesic effect on rats with incision pain.It is feasible for Xuelian injection to replace tramadol as an analgesia option for moderate and severe pain after surgery.
