摘要
目的探究振腹治疗寒凝证类痛经大鼠的优效参数。方法采用全身冷冻法结合苯甲酸雌二醇、缩宫素注射法建立寒凝痛经大鼠模型,随机分为盐水组、模型组以及9个振腹组,共11个组,每组6只。在冷冻5天、振腹干预5天,同时每日注射苯甲酸雌二醇致敏后,注射缩宫素致痛,立即测定大鼠扭体潜伏期、扭体次数和子宫体表投影温度,并计算扭体评分。(1)采用方差分析法观察盐水组、模型组和9个振腹组间的差异。(2)采用三因素三水平正交实验方差分析9个振腹组的行为学结果,找出影响振腹疗效的主因素,并得出疗效最优参数组合。结果(1)与盐水组比较,模型组扭体潜伏期明显缩短,扭体评分明显增加,子宫体表投影温度明显降低(P<0.01);与模型组比较,4个振腹组扭体潜伏期明显延长(P<0.05),8个振腹组扭体评分明显减少(P<0.05),所有振腹组子宫体表投影温度均显著增高(P<0.05)。(2)振腹组正交实验结果显示:振腹疗法最主要的影响因素为治疗时间(P<0.01)。综合考虑痛经发作潜伏期、疼痛程度及子宫温度,振腹治疗寒凝证类痛经大鼠的综合优效参数组合为长时间、高频、低幅。结论振腹疗法能够延长寒凝证痛经大鼠的潜伏期,减轻疼痛,提高子宫温度。治疗时间是影响振腹法治疗寒凝证类痛经的最主要参数。长时间、高频、低幅振动是治疗寒凝证类痛经的最优效参数组合。
Objective To explore the optimal parameters of primary dysmenorrhea rats with coagulated cold syndrome treated with vibrating abdomen.Methods The primary dysmenorrhea rat model with coagulated cold syndrome was established by whole body freezing method combined with estradiol benzoate and oxytocin injection.The rats were randomly divided into saline group,model group and 9 vibration groups,a total of 11 groups,6 in each group.After freezing for 5 days,vibrating abdomen intervention for 5 days,and daily injection of estradiol benzoate to sensitize the rats,oxytocin was injected to induce pain.The incubation period of torsion,the number of torsion and the temperature of uterine surface were measured immediately,and the torsion score was calculated.(1)The ANOVA was used to observe the differences among the saline group,the model group and the 9 vibrating abdomen groups.(2)The ANOVA of three factors and three levels of orthogonal experiment were used to analyze the behavioral results of 9 vibrating abdomen groups,to find out the main factors affecting the efficacy of vibrating abdomen,and to obtain the optimal combination of parameters.Results(1)Compared with the saline group,the incubation period of torsion in the model group was significantly shortened,the torsion score was significantly increased,and the projection temperature of uterine body surface was significantly decreased(P<0.01).Compared with the model group,the incubation period of torsion was significantly prolonged in the 4 four vibrating abdomen groups(P<0.05),the score of torsion was significantly reduced in the eight vibrating abdomen groups(P<0.05),and the surface projection temperature of uterus in all vibrating abdomen groups was significantly increased.(2)The results of orthogonal experiment in the vibrating abdomen group showed that the most important factor affecting the vibrating abdomen therapy was the treatment time(P<0.01).Considering the preclinical period of dysmenorrhea,the degree of pain,and the temperature of uterus,the comprehensive optimal parameter combination of dysmenorrhea rats with coagulated cold syndrome treated with vibrating abdomen method,was long time,high frequency and low amplitude.Conclusion Vibrating abdomen therapy can prolong the incubation period,relieve pain and raise the temperature of uterus of dysmenorrhea rats with coagulated cold syndrome.Treatment time is the most important parameter affecting dysmenorrhea with coagulated cold syndrome.Long time,high frequency and low amplitude vibration are the best combination of effective parameters for the treatment of dysmenorrhea with coagulated cold syndrome.
