Correlation between TRAIL and resistance-related genes in patients with epithelial ovarian cancer

摘要 目的探讨TRAIL与上皮性卵巢癌耐药相关基因的关系,筛选出和TRAIL相关的耐药因子,从而为预测卵巢癌的耐药并指导临床用药提供参考。方法选取50例上皮性卵巢癌患者作为试验组,10例良性卵巢上皮性肿瘤患者为对照组,采用免疫组化方法检测TRAIL在上皮性卵巢癌组织中的表达情况。分析相应组织切片中MDR1、LIVIN、TOPOⅠ、TOPOⅡ、PTEN、ERCC1耐药相关蛋白的表达。结果TRAIL与MDR1表达呈负相关(r=-1;P=0);TRAIL与LIVIN、TOPOⅠ、TOPOⅡ、PTEN、ERCC1的表达无相关性(r值分别为0.105、-0.8、-0.798、-0.861和-0.01,P>0.05)。结论TRAIL和MDR1的表达呈负相关,和LIVIN、PTEN、TOP0Ⅰ、TOPOⅡ、ERCC1的表达无明显相关性。TRAIL或许可以通过降低耐药因子MDR1的表达来改变卵巢癌的多药耐药。 Objective To investigate the correlation between TRAIL and drug resistance-related proteins including MDR1,LIVIN,TOPOⅠ,TOPOⅡ,PTEN and ERCC1 in patients with epithelial ovarian cancer,so as to provide reference for predicting the drug resistance of ovarian cancer and to guide clinical medication.Methods Fifty patients with epithelial ovarian cancer were enrolled as experimental group,and the other 10 patients with benign epithelial ovarian tumor were enrolled as control group.The expression levels of TRAIL in epithelial ovarian cancer tissues were detected by immunohistochemistry.The expression levels of drug resistance-related proteins including MDR1,LIVIN,TOPOⅠ,TOPOⅡ,PTEN and ERCC1 in corresponding tissues were observed.Results The expression levels of TRAIL were negatively correlated with those of MDR1(P<0.01).There were no significant differences in the expression between TRAIL and LIVIN,TOPOⅠ,TOPOⅡ,PTEN,ERCC1(P>0.05).Conclusion The expression levels of TRAIL are negatively correlated with those of MDR1,however,there are no significant differences in the expression between TRAIL and LIVIN,TOPOⅠ,TOPOⅡ,PTEN,ERCC1.TRAIL may change the multidrug resistance in patients with epithelial ovarian cancer by decrease the expression levels of MDR1.
