摘要
目的观察温阳振衰颗粒对阿霉素诱导性慢性心力衰竭(CHF)模型兔的心肌Toll样受体2(TLR2)和核转录因子-κB(NF-κB)蛋白表达的影响。方法耳缘静脉注射阿霉素构建CHF兔模型。按心功能分层将造模成功兔随机分为5组:模型组、对照组和低、中、高3个剂量实验组,每组11只。另取11只正常兔作为正常组。在第5周开始,低、中、高3个剂量实验组分别予温阳振衰颗粒0.4,0.8,1.6 g·kg^-1用蒸馏水稀释后灌胃;对照组予以依那普利5 mg·kg^-1配以蒸馏水灌胃,正常组和模型组均灌服等量蒸馏水,每日1次,均连续灌胃4周。用蛋白质印迹法检测心肌TLR2和NF-κB蛋白的表达。结果正常组、模型组、对照组和低、中、高3个剂量实验组的TLR2蛋白相对表达量(灰度值比值)分别为0.74±0.06,0.90±0.02,0.78±0.02,0.81±0.03,0.80±0.02和0.78±0.02;这6组的NF-κB蛋白相对表达量(灰度值比值)分别为0.43±0.02,0.71±0.02,0.51±0.02,0.64±0.02,0.60±0.05和0.52±0.03。以上指标:模型组与正常组比较,差异均有统计学意义(均P<0.01);低、中、高3个剂量实验组和对照组与模型组比较,差异均有统计学意义(均P<0.05)。结论温阳振衰颗粒可降低心力衰竭模型兔心肌TLR2及NF-κB蛋白的表达,这可能是其治疗心力衰竭时抑制细胞炎症反应的作用机制之一。
Objective To observe the effects of Wenyangzhenshuai Granules on the expression of Toll-like receptor 2(TLR2)and nuclear factor kappa B(NF-κB)of the myocardial tissue in the rabbits with chronic heart failure(CHF)induced by adriamycin.Methods The CHF model was established by intravenous injection of adriamycin at ear margin.The CHF model rabbits were randomly divided into five groups:model group,control group and experimental-L,experimental-M and experimental-H groups,each group had 11 rabbits.The other 11 rabbits were not injected with adriamycin as normal group.The normal group and model group were given distilled water by gavage;control group was given enalapril 5 mg·kg^-1 by gavage;experimental-L,experimental-M and experimental-H groups were respectively administered with Wenyangzhenshuai Granules 0.4,0.8 and 1.6 g·kg^-1,once daily for 4 weeks.Western blot was used to detect the expression of TLR2 and NF-κB in myocardium.Results The relative expression levels(ratio of gray value)of TLR2 protein in normal group,model group,control group,and experimental-L,experimental-M,experimental-H groups were 0.74±0.06,0.90±0.02,0.78±0.02,0.81±0.03,0.80±0.02,0.78±0.02;the relative expression levels(ratio of gray value)of NF-κB protein in the 6 groups were 0.43±0.02,0.71±0.02,0.51±0.02,0.64±0.02,0.60±0.05,0.52±0.03.Comparison between model group and normal group,the difference of the factors were significantly(all P<0.01);comparison between three dose experimental groups and control group with model group,the differenceof the factors were significantly(all P<0.05).Conclusion Wenyangzhenshuai Granules can reduce the expression of TLR2 and NF-κB protein in myocardium of rabbits with CHF,which may be one of the mechanisms of its role in inhibiting cellular inflammatory response in the treatment of heart failure.
