绞股蓝总皂苷对非酒精性脂肪肝大鼠氧化应激及肝细胞凋亡的影响 被引量:19

Effect of gynostemma pentaphyllum on oxidative stress and liver cell apoptosis of nonalcoholic fatty liver disease rats

摘要 目的观察绞股蓝总皂苷对非酒精性脂肪肝大鼠氧化应激及肝细胞凋亡的影响。方法将30只建模成功的非酒精性脂肪肝大鼠随机分为模型组与实验组,各15只;另随机选取15只SD大鼠正常饲养并作为对照组,实验组大鼠灌胃300 mg·kg^-1绞股蓝总皂苷,模型组及对照组大鼠灌胃等量生理盐水,各组均连续干预12周。以全自动生化分析仪检测大鼠肝功能指标和血脂水平;以黄嘌呤氧化酶法检测超氧化物歧化酶(SOD)含量,以硫代巴比妥比色法检测丙二醛(MDA)含量;以苏木精-伊红(HE)染色检测大鼠肝组织病理学形态;以TUNEL法检测大鼠肝细胞凋亡情况。结果对照组、模型组及实验组大鼠血清三酰甘油(TG)含量分别(1.41±0.42),(3.21±0.67),(2.12±0.38)mmol·L^-1;胆固醇(TC)含量分别为(1.13±0.39),(2.84±0.56),(1.46±0.42)mmol·L^-1;高密度脂蛋白胆固醇(HDL-C)含量分别为(0.31±0.09),(2.03±1.01),(0.55±0.04)mmol·L^-1;低密度脂蛋白胆固醇(LDL-C)含量分别为(0.91±0.10),(0.22±0.03),(0.65±0.04)mmol·L^-1;血清丙二醛(MDA)含量分别为(10.03±2.14),(18.41±2.26),(13.25±1.94)nmol·mL^-1;血清超氧化物歧化酶(SOD)含量分别为(362.73±24.55),(251.06±20.31),(276.81±17.99)U·mL^-1;肝指数分别为(2.09±0.46)%,(3.56±0.67)%,(2.61±0.55)%;凋亡指数分别为(2.01±0.43)%,(31.25±1.97)%,(8.12±0.94)%;与对照组比较,模型组血清GOT、GPT、MDA含量及肝指数、肝细胞凋亡指数升高,而实验组较模型组降低;模型组大鼠血清SOD含量较对照组降低,而实验组较模型组升高,且以上指标各组间差异均有统计学意义(均P<0.05)。结论绞股蓝总皂苷可有效抑制非酒精性脂肪肝大鼠氧化应激反应及肝细胞凋亡,恢复其肝功能。 Objective To explore the effect of gynostemma pentaphyllum on oxidative stress and liver cell apoptosis of nonalcoholic fatty liver disease rats.Methods Thirty nonalcoholic fatty liver disease rats after modeling successfully were randomly divided into model group and test group,with 15 rats in each group;another 15 SD rats with normal breeding were selected as control group.Rats in test group were gavage with 300 mg·kg^-1 gynostemma pentaphyllum,rats in model group and control group were gavage with the same amount of normal saline,each group was intervented for 12 weeks.The liver function,blood lipid indexes of rats were detected by automatic biochemical analyzer;superoxide dismutase(SOD)contents were detected by xanthine oxidase method,malondialdehyde(MDA)contents were detected by glucosinolates barbitone colorimetry method;the rat liver tissue pathological damage was detected by hematoxylin and eosin(HE)staining;the liver cell apoptosis were detected by Terminal deoxynucleotidyl transferase d UTP nick-end labeling(TUNEL).Results Serum triglyceride(TG)in control group,model group and test group were(1.41±0.42),(3.21±0.67),(2.12±0.38)mmol·L^-1 respectively;total cholesterol(TC)were(1.13±0.39),(2.84±0.56),(1.46±0.42)mmol·L^-1 respectively;high density lipoprotein cholesterol(HDL-C)were(0.31±0.09),(2.03±1.01),(0.55±0.04)mmol·L^-1 respectively;low density lipoprotein cholesterol(LDL-C)were(0.91±0.10),(0.22±0.03),(0.65±0.04)mmol·L^-1;serum malondialdehyde(MDA)were(10.03±2.14),(18.41±2.26),(13.25±1.94)nmol·m L^-1;serum superoxide dismutase(SOD)were(362.73±24.55),(251.06±20.31),(276.81±17.99)U·m L^-1;liver indexes were(2.09±0.46)%,(3.56±0.67)%,(2.61±0.55)%;liver cell apoptosis indexes were(2.01±0.43)%,(31.25±1.97)%,(8.12±0.94)%.Compared with control group,serum GOT,GPT,MDA contents and liver index,liver cell apoptosis were increased in model group,while test group were lower than model group;serum SOD content in model group was lower than that of control group,while test group was higher than that of model group,and the above indexes in each group had significant difference(all P<0.05).Conclusion Gynostemma pentaphyllum can inhibit the oxidative stress reaction and liver cell apoptosis of nonalcoholic fatty liver disease rats effectively,restore its function of liver.
