摘要
目的研究替普瑞酮对慢性萎缩性胃炎大鼠胃黏膜组织环氧合酶-2(COX-2)表达的影响。方法将28只慢性萎缩性胃炎模型大鼠随机分为模型组及实验组,各14只,另随机选取15只正常大鼠为对照组,实验组灌胃替普瑞酮200 mg·kg^-1·d^-1,对照组及模型组均灌胃等量生理盐水,每天1次,连续干预4周。以酶联免疫吸附(ELISA)法检测血清细胞因子含量;以苏木精-伊红(HE)染色观察大鼠胃黏膜组织病理学形态;以免疫组化法检测胃黏膜组织COX-2蛋白表达。结果对照组、模型组和实验组大鼠血清白细胞介素-1β(IL-1β)含量分别为(42.09±8.23),(80.04±10.06),(62.11±5.74)ng·L^-1;白细胞介素-6(IL-6)含量分别为(10.21±0.41),(96.77±19.78),(63.46±10.24)pg·mL^-1;肿瘤坏死因子-α(TNF-α)分别为(7.04±2.04),(79.19±12.41),(35.61±9.16)pg·mL^-1;血清COX-2蛋白表达量分别为(2.14±0.21),(16.37±2.14),(7.53±1.06)ng·mL^-1;胃黏膜组织COX-2蛋白阳性细胞百分比分别为(14.68±3.14)%,(42.79±6.33)%,(25.89±4.01)%。模型组上述指标均较对照组升高,而实验组均较模型组降低(均P<0.05)。结论替普瑞酮可有效抑制慢性萎缩性胃炎大鼠胃黏膜组织COX-2表达,改善其胃黏膜炎性病理损伤,修复其黏膜屏障。
Objective To explore the effect of teprenone on the gastric mucosa tissue cyclooxygenase-2(COX-2)expression of chronic atrophic gastritis rats.Methods Twenty-eight chronic atrophic gastritis model rats were randomly divided into model group and test group,14 rats in each group,another 15 normal rats were selected as control group.Test group was gavage with 200 mg·kg^-1·d^-1 teprenone,control group and model group were gavage with the same amount of normal saline,1 time·d^-1,continuously intervened for 4 weeks.The serum cytokine levels were detected by enzyme-linked immunosorbent assay(ELISA),the gastric mucosa tissue pathology morphology were observed by hematoxylin and eosin(HE)staining;the gastric mucosa tissue COX-2 protein expression were detected by immunohistochemical method.Results The serum interleukin-1β(IL-1β)in control group,model group,test group were(42.09±8.23),(80.04±10.06),(62.11±5.74)ng·L^-1;the interleukin-6(IL-6)were(10.21±0.41),(96.77±19.78),(63.46±10.24)pg·m L^-1;the tumor necrosis factor-α(TNF-α)were(7.04±2.04),(79.19±12.41),(35.61±9.16)pg·m L^-1;the serum COX-2 protein expression were(2.14±0.21),(16.37±2.14),(7.53±1.06)ng·m L^-1;the gastric mucosa tissue COX-2 protein positive cells proportion were(14.68±3.14)%,(42.79±6.33)%,(25.89±4.01)%.The above indexes in model group were higher than those in control group,while test group were lower than model group(all P<0.05).Conclusion Tipridone can inhibit the gastric mucosa tissue COX-2 protein expression effectively,and improve the gastric mucosa pathological damage,repair the mucosal barrier.
