隔姜灸天枢穴对UC大鼠结肠组织的mi RNA表达谱的影响 被引量:8

Effect of Ginger-partitioned Moxibustion at Tianshu(ST25)on the miRNA Expression Profile in Colon Tissues of Ulcerative Colitis Rats

摘要 目的探讨隔姜灸天枢穴对葡聚糖硫酸钠(DSS)诱导的溃疡性结肠炎(UC)大鼠结肠组织mi RNA表达谱的影响,从mi RNA的角度探讨隔姜灸的效应机制。方法将30只健康成年SD雄性大鼠随机分为正常组(NC组)、模型组(UC组)和隔姜灸组(GM组),每组10只。采用4%DSS制备UC大鼠模型,GM组取双侧天枢穴进行隔姜灸干预。干预7 d后,采用苏木精-伊红染色观察隔姜灸干预UC大鼠的效应,并采用RNA-seq的方法检测大鼠结肠组织的miRNA表达谱。结果病理结果提示,隔姜灸能明显减轻UC大鼠结肠组织的炎症。与NC组比较,UC组大鼠结肠组织差异表达mi RNAs有45个,其中上调42个,下调3个;与UC组比较,GM组大鼠结肠组织差异表达mi RNAs有8个,其中上调3个,下调5个。UC病理状态下表达量改变的mi R-139-5p,miR-204和miR-184,均可被隔姜灸天枢穴干预后逆转,其预测靶基因的pathways与UC的免疫炎症密切相关。结论隔姜灸天枢穴对DSS诱导的UC大鼠结肠组织黏膜损伤具有保护效应,这与其影响结肠组织miRNA表达谱相关,主要调节了UC结肠组织异常表达的miR-139-5p,miR-204和mi R-184。 Objective To study the effect of ginger-partitioned moxibustion at Tianshu(ST25)on the miRNA expression profile in colon tissues of Dextran sulfate sodium(DSS)-induced ulcerative colitis(UC)rat models,for discussing the action mechanism of ginger-partitioned moxibustion from the perspective of miRNA.Method Thirty healthy adult male Sprague-Dawley(SD)rats were randomized into a normal control group(NC group),a model group(UC group)and a ginger-partitioned moxibustion group(GM group),with 10 rats in each group.UC rat model was developed using 4%DSS,and the GM group was intervened by ginger-partitioned moxibustion at bilateral Tianshu.After 7-day intervention,the effect of ginger-partitioned moxibustion in intervening UC rats was observed using Hematoxylin-eosin(HE)staining,and the miRNA expression profile in rat colon tissues was detected by RNA-seq technique.Result The pathological results showed that ginger-partitioned moxibustion significantly reduced inflammation in colon tissues of UC rats.Compared with the NC group,there were 45 miRNAs differentially expressed in rat colon tissues in the UC group,of which,42 were up-regulated and 3 were down-regulated;compared with the UC group,there were 8 miRNAs differentially expressed in rat colon tissues in the GM group,of which,3 were up-regulated and 5 were down-regulated.The expression levels of miR-139-5p,miR-204 and miR-184,which changed under UC pathological condition,all can be reversed after intervention of ginger-partitioned moxibustion at Tianshu,and the pathways of their predicted target genes were closely related to immune inflammation.Conclusion Ginger-partitioned moxibustion at Tianshu can produce protective effect on colonic mucosa injury in DSS-induced UC rats,which is related to its influence on the miRNA expression profiles in colon tissues,mainly manifested by the regulation of the abnormal expressions of miR-139-5p,miR-204 and miR-184.
