Determination of 17 Amino Acids in Source Water by UPLC-MS/MS and Its Application

摘要 氨基酸是水中重要的含氮有机物,也是部分含氮消毒副产物的前体物。为了解国内实际水源水中溶解性氨基酸的存在情况,建立了超高效液相色谱串联质谱(UPLC-MS/MS)直接测定水中17种氨基酸的检测方法,并对不同流域水源水中的溶解态氨基酸存在情况进行了研究。该方法的线性范围为1~200 nmol/L,检出限为0.2~1.5 nmol/L,相关系数(R2)均大于0.99,方法精密度在0.11%~4.66%之间,加标回收率在88.31%~114.92%之间,满足水环境中氨基酸分析的要求,为地表水的氨基酸浓度检测提供了简便、可靠的方案。采用该方法检测实际样品的结果表明,长江、汉江、东江和千岛湖水中溶解性氨基酸总浓度分别为1296、1348、885和55 nmol/L。 Amino acids are important part of nitrogen-containing organic matters in water and precursors of nitrogen-containing disinfection by-products.In order to understand the existence of dissolved amino acids in source water of China,a method for determining 17 amino acids in water by ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)was established,and the occurrence of amino acids in different source waters was studied.The results showed that the linear range of the method was 1-200 nmol/L,the detection limit was 0.2-1.5 nmol/L,the coefficient of correlation(R2)was greater than 0.99,the precision of the method was from 0.11%to 4.66%,and the recovery ratio of the spiking samples were from 88.31%to 114.92%,which could meet the requirements of amino acids detection in water environment.This method provided a simple and reliable plan for the detection of amino acid concentration in surface water.The results of the actual sample showed that the total concentrations of dissolved amino acids in the Yangtze,Hanjiang,Dongjiang and Qiandao Lake waters were 1296 nmol/L,1348 nmol/L,885 nmol/L and 55 nmol/L,respectively.
