摘要
目的探讨低氧诱导的大鼠脂肪干细胞旁分泌因子对子痫前期大鼠心肌损伤的保护作用。方法从未孕雌鼠分离并培养获得第3代脂肪干细胞,通过流式细胞仪对5种表面标志物(CD73、CD90、CD14、CD19、CD34)进行鉴定。将第3代脂肪干细胞进行控制性低氧培养制备脂肪干细胞上清液,采用ELISA法检测上清液中相关细胞因子[表皮生长因子(EGF)、胰岛素样生长因子(IGF)、碱性成纤维细胞生长因子(bFGF)、转化生长因子(TGF-α)]的水平。将21只孕鼠分为3组(造模组,造模+治疗组和对照组),用亚硝基左旋精氨酸甲酯(L-NAME)建立子痫前期模型,造模+治疗组注射干细胞上清液,造模组和对照组注射生理盐水。测定孕鼠心肌损伤标志物[肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)、肌钙蛋白T(cTnT)、天冬氨酸转氨酶(AST)、心脏脂肪酸结合蛋白(H-FABP)]水平,心肌钙蛋白酶活性和相关基因表达量,并采用HE染色观察3组心肌组织形态。结果流式细胞仪鉴定第3代脂肪干细胞CD73、CD90阳性,CD14、CD19、CD34阴性。上清液中EGF、IGF、bFGF、TGF-α水平较高。造模组CK-MB、LDH、cTnT、AST、H-FABP水平均高于对照组和造模+治疗组(P<0.05或0.01)。钙蛋白酶活性和Capn1基因表达量造模组高于造模+治疗组,造模+治疗组高于对照组(均P<0.05)。HE染色提示造模+治疗组的心肌组织横纹较造模组清晰,细胞异常程度较轻。结论低氧诱导的脂肪干细胞旁分泌因子能够减轻子痫前期所致的心肌损伤。
Objective To investigate the protective effect of hypoxia-induced paracrine cytokines of adiposederived stem cells on myocardial injury in preeclampsia rats.Methods Adipose-derived stem cells were isolated from non-pregnant female rats and cultured to the third generation.Five cell surface markers(CD73、CD90、CD14、CD19、CD34)were identified by flow cytometer analysis.The third generation adipose stem cells were cultured under controlled hypoxia to prepare the supernatant of adipose stem cells.The contents of related cytokines[epidermal growth factor(EGF),insulin-like growth factor(IGF),basic fibroblast growth factor(bFGF),transforming growth factor(TGF-α)]in the supernatant were detected by ELISA.21 pregnant rats were divided into 3 groups(model group,model+treatment group and control group).The model of preeclampsia was established with nitroso-L-arginine methyl ester(L-NAME).The model+treatment group were injected with stem cell supernatant,and the model group and control group were injected with normal saline.The contents of myocardial injury markers[creatine kinase isoenzyme(CK-MB),lactate dehydrogenase(LDH),troponin T(cTnT),aspartate aminotransferase(AST),cardiac fatty acid binding protein(H-FABP)],the calpain activity and the expression of related genes were measured.the myocardial morphology of the three groups was observed by hematoxylin-eosin staining.Results Flow cytometry showed that the third generation of adipose stem cells were positive for CD73 and CD90,but negative for CD14,CD19 and CD34.The contents of EGF,IGF,bFGF and TGF-αin the supernatant were high.The contents of CK-MB,LDH,cTnT,AST and H-FABP in the model group were higher than those in the control group and the model+treatment group(P<0.05 or 0.01).Calpain activity and the expression of Capn1 gene in the model group were higher than those in the model+treatment group,and those in the model+treatment group were higher than those in the control group(all P<0.05).HE staining showed that the transverse striations of myocardial tissue in the model+treatment group were clearer and the degree of cell abnormality was slighter than that in the model group.Conclusion Hypoxic-induced paracrine cytokines of adipose-derived stem cells can reduce cardiac injury caused by preeclampsia.
