三七皂苷R1的大鼠血浆药物代谢动力学研究

摘要 目的通过测定三七皂苷血浆浓度随时间变化的情况,研究其在生物体内的代谢和运动过程。方法D大鼠服用三七皂苷R 1提取液后,于0~24h内按时间点取血,血浆经乙腈沉淀蛋白处理后,所得供试品溶液注入高效液相色谱仪中分析,使用Agilent SB C 18(250×4.6mm,5μm)色谱柱,流动相采用乙腈与水按一定比例混合,设置梯度程序洗脱,流速1.0mL·min^-1,进样量10μL,检测波长203nm,共测定6只SD大鼠的血样,记录峰面积,根据标准曲线计算血浆中待测物的含量,之后将含量数据输入DAS 2.1软件中计算药代动力学参数。结果三七皂苷R 1的T max为1.0h,C max为(537.26±50.11)μg·mL^-1,T 1/2为(9.18±1.45)h,MRT为(8.86±1.90)h,AUC i为(1678.54±147.31)h/(μg·mL),AUC∞为(2031.64±220.15)h/(μg·mL)。结论本研究快速、准确、高效,能够较为全面地对三七皂苷R 1的体内运动特征进行研究。 OBJECTIVE Through determination of plasma concentration of noto ginseng saponin R 1 and its change by the time,to investigate its metabolism and dynamics in animal body.METHODS extract containing panax no toginseng saponin R 1 was given to the SD rat,followed by blood taking within 24h,the obtained plasma was precipitated to remove protein by acetonitrile,and the sample solutions were injected to high performance chromatography for analy sis;in which aAgilent SBC 18(5μm,250×4.6mm)column was used,with a mobile phase of acetonitrile-water mixture in certain ratios,and a gradient pr ogram was set for separation,the flow rate was 1.0mL·min^-1,the injecti on volume was 10μL,the detection wavelength was 203nm,6 SD rats were used for experiment,and the plasma concentration of panax notoginseng saponin R 1 was calculated by DAS 2.1 software for pharmacokinetic parameters.RESULTS The mean values for panax notoginseng saponin R 1 were as follows:T max=1.0h,C max=(537.26±50.11)μg·mL^-1,T 1/2=(9.18±1.45)h,MRT=(8.86±1.90)h,AUC i=(1678.54±147.31)h/(μg·mL),AUC∞=(2031.64±220.15)h/(μg·mL).CONCLUSION This study develo an analytical m ethod ofquickness,accuracy and efficiency,which was capable of investigating in body dynamics of panax notoginseng saponin R1 comprehensively.
