摘要
目的研究饮片混合煎煮与配方颗粒调制的干姜白术半夏汤在实际临床用量上是否存在差异。方法通过测定两种方法制备的干姜白术半夏汤中6-姜辣素和白术内酯Ⅰ的含量,考察是否存在显著性差异。含量测定采用高效液相色谱法,使用Wa ters Xbridge色谱柱(250×5mm,4.6μm),乙腈-0.2%磷酸为流动相,梯度洗脱,流速1.0mL·min^-1,检测波长254nm,柱温30℃,进样量10μL;共测定10批饮片及配方颗粒调制的汤剂。结果10批配方颗粒调制的样品中,6-姜辣素的含量为0.0453~0.0587mg·mL^-1,相当于饮片煎煮样品的105.79~155.10%;白术内酯Ⅰ的含量为0.0246~0.0317mg·mL^-1,相当于饮片煎煮样品的86.12%~124.10%。结论配方颗粒调制干姜白术半夏汤中,6-姜辣素含量显著高于饮片煎煮制备的汤剂,白术内酯Ⅰ的含量则无显著性差异,提示配方颗粒可以替代饮片用于制备干姜半夏白术汤。
OBJECTIVE To investigate the clinical usage differences of Ganjiang Baizhu Banxia decoction prepared by decoction pieces and TCM formula granule.METHODS The differences of the two above mentioned preparati on method was performed by determination of 6-gingerol and atractylodes lactoneⅠ,analysis was carried out by HPLC,the column used was Waters Xbridge(250×5 mm,4.6μm),acetonitrile-0.2%phosphoric acid as mobile phase,the elution mode was gradient,the flow rate was 1.0mL·min^-1,the detection wavelength was 254nm,the column temperature was 30℃and the injection volume was 10μL;10 batches of decoction were prepared by TCM formula granule,as well as by co rresponding decoction pieces.RESULTS in 10 batches of the samples,contents of 6-gingerol were 0.0453-0.0587mg·mL^-1,equivalent to 105.79-155.10%of the ones prepared by decoction pieces;the values for that of atractylodes lactoneⅠwere 0.0246-0.0317mg·mL^-1 and 86.12-124.10%,respectively;CONCLUSION Ganjiang Baizhu Banxia decoction pr epared by TCM formula granule has a significant higher 6-gingerol content,while the atractylodes lactoneⅠcontent shows not difference between the two methods,the above mentioned results indicates that decoction pieces can be replaced by TCM formula granule in preparation of Ganjiang Baizhu Banxia decoction.
