骨刺散对热板法及甲醛法致小鼠疼痛的影响 被引量:5

The Influence of Bony Spur Powder on Mice with Pain Induced by Hot Plate Method and Formaldehyde Method

摘要 目的:观察骨刺散对热板法、甲醛法致小鼠疼痛的影响。方法:分别将实验小鼠按照随机数字表法分为空白组、白醋组、骨质宁擦剂组、骨刺散大剂量组和骨刺散小剂量组,每组各10只。热板法实验中空白组给予生理盐水湿敷,每足0.3 mL;白醋组给予白醋湿敷,每足0.3 mL;骨质宁擦剂组给予骨质宁擦剂湿敷,每足0.3 mL;骨刺散大剂量组将0.5 g骨刺散溶解于1 mL白醋给予小鼠后足用药,厚度2 mm;骨刺散小剂量组将0.25 g骨刺散溶解于1 mL白醋给予小鼠后足用药,厚度2 mm。甲醛法实验给药方法同热板法,给药后立即在小鼠右后足背皮下注射新配制的体积分数2.5%甲醛溶液30μL。测定给药前、给药后30 min、给药后60 min、给药后90 min在热板仪中再次测定小鼠痛阈,观察并记录每只小鼠首次舔咬右后足的潜伏期及5 min内舔咬右后足的次数和10 min内舔咬右后足的次数。结果:热板法实验结果显示:与空白组比较,骨刺散大剂量组、骨刺散小剂量组和骨质宁擦剂组在给药后30 min、给药后60 min、给药后90 min时,可显著提高小鼠痛阈值,差异均有统计学意义(P<0.05)。甲醛法实验结果显示:与空白组比较,白醋组、骨刺散大剂量组、骨刺散小剂量组及骨质宁擦剂组可延长小鼠首次舔右后足潜伏期,同时可减少小鼠5 min和10 min内的舔足次数,差异具有统计学意义(P<0.05)。结论:骨刺散有较好的镇痛作用,可显著改善小鼠由于软组织损伤、外伤导致的肿胀疼痛。 Objective:To observe the influence of Bony Spur Powder on mice with pain induced by hot plate method and formaldehyde method.Methods:The experimental mice were divided into the blank group,the white vinegar group,the Guzhining(骨质宁,GZN) liniment group,the Bony Spur Powder high-dose group and the Bony Spur Powder low-dose group according to the random number table method,with 10 mice in each group.The blank group was given 0.3 mL of normal saline for wet compress.And the white vinegar group was given 0.3 mL of white vinegar for wet compress.The GZN liniment group was given 0.3 mL of GZN liniment for wet compress.In the high-dose group,0.5 g of Bony Spur Powder was dissolved in 1 mL of white vinegar and then given to the hind foot of mice,with a thickness of 2 mm.In the high-dose group,0.25 g of Bony Spur Powder was dissolved in 1 mL of white vinegar and then given to the hind foot of mice,with a thickness of 2 mm.In the formaldehyde method experiment,30 μ L of newly prepared 2.5% formaldehyde solution was injected subcutaneously into the back of right hind foot of mice immediately after administration.Before administration,30 minutes after administration,60 minutes after administration and 90 minutes after administration,the pain threshold of mice was measured again respectively in the hot plate apparatus.The incubation period of the first licking and biting of the right hind foot of each mouse,the times of licking and biting the right hind foot within 5 minutes and the times of licking and biting the right hind foot within 10 minutes were observed and recorded.Results:The results of hot plate method showed that:compared with the blank group,the pain threshold of mice in the groups of high-dose Bony Spur Powder,low-dose Bony Spur Powder and GZN liniment could be significantly increased 30 minutes,60 minutes and 90 minutes after administration,and all the differences were statistically significant(P<0.05).The results of formaldehyde test showed that:compared with the blank group,the white vinegar group,the groups of high-dose Bony Spur Powder,low-dose Bony Spur Powder and GZN liniment could prolong the incubation period of the first licking of the right hind foot,and reduce the times of licking the foot within 5 minutes and 10 minutes,and all the differences were statistically significant(P<0.05).Conclusion:Bony Spur Powder has a good analgesic effect and can significantly improve the swelling and pain of mice caused by soft tissue injury and trauma.
