HPLC梯度洗脱法测定甲磺酸二氢麦角毒碱缓释片的含量

HPLC Gradient Elution Method for the Determination of Dihydroergotoxine Methanesulfonate Sustained Release Tablets

目的采用HPLC梯度洗脱法测定甲磺酸二氢麦角毒碱缓释片的含量。方法采用Welch C18柱(250mm×4.6mm,5μm),梯度洗脱流动相:醋酸铵溶液-乙腈;流速为1.0mL·min^-1,检测波长为280nm,柱温为30℃,进样量20μL。结果甲磺酸二氢麦角毒碱缓释片中四组分能有效分离,在9.408~94.08μg·mL^-1浓度范围内四组分及总成分的线性关系均良好(r=0.9997)。总碱平均回收率分别为100.58%、99.42%、99.43%(n=9)。结论该法简便、准确、专属性好,可以用于甲磺酸二氢麦角毒碱缓释片的含量测定。

OBJECTIVE HPLC gradient elution method was established to determine the content of Dihydroergotoxine Methanesulfonate sustained release tablets.METHODS The HPLC method was performed on a Welch C18 column(250 mm×4.6 mm,5μm).Acetonitrile and ammonium acetate were used as the gradient elution mobile phase.The flow rate was 1.0 mL·min^-1.The detection wavelength was 280 nm and the column temperature was 30℃.The injection volume was 20μL.RESULTS The four components in the dihydroergotoxine methanesulfonate sustained release tablets were basically separated.The four components and the total component were showed good linearity at 9.408-94.08μg·mL^-1(r=0.9997).The average recoveries were 100.58%,99.42%,99.43%(n=9),respectively.CONCLUSION The method is simple,accurate and specific.It can be used for the determination of the content of Dihydroergotoxine Methanesulfonate sustained release tablets.
