化脓隐秘杆菌候选抗原的免疫保护效果测试

Immunoprotection Effect Testing of A Candidate Antigen of Trueperella pyogenes

为鉴定化脓隐秘杆菌的新抗原,对被抗血清识别的化脓隐秘杆菌膜蛋白进行质谱分析,采用免疫印迹分析抗原性,ELISA方法测试被免疫小鼠的抗体水平,通过化脓隐秘杆菌攻毒测试免疫保护率。结果显示,铁ABC(ATP结合盒,ATP binding cassette)转运体的底物结合蛋白是化脓隐秘杆菌的1个新保护性抗原,具有良好的抗原性和免疫原性。

In order to identify new antigens of Trueperella pyogenes,a membrane protein of T.pyogenes recognized by the antiserum was identified by using mass spectrometry,and its antigenicity was analyzed by immunoblotting.Antibody level of mice immunized by the membrane protein was tested by using ELISA,and the immune protection rate for the immune mice was evaluated by challenged with T.pyogenes.The results showed that substrate binding protein of iron ABC(ATP binding cassette)transporter was a new protective antigen of T.pyogenes,and it had good antigenicity and immunogenicity.
