摘要
目的分析FAM96A在乳腺癌组织中表达及其对乳腺癌细胞增殖凋亡和侵袭转移的影响。方法收集的人乳腺癌组织及癌旁组织标本75例,采用免疫印迹法(W-B)法检测乳腺癌组织和癌旁组织中的FAM96A表达水平,并以人乳腺癌MCF-7细胞作为研究对象,分为2组分别转染pcDNA3.1-vector空白质粒和pcDNA3.1-myc-FAM96A质粒,对比两组MCF-7细胞活性、凋亡率及侵袭转移的差异。结果乳腺癌组织中FAM96A蛋白表达水平为(0.31±0.05)较癌旁组织(1.82±0.27)明显降低;过表达FAM96A后72 h、96 h时,细胞活性较对照组明显减弱(均P<0.05);过表达FAM96A后的细胞凋亡率升高[(32.19±1.02)%比(3.07±0.84)%],穿膜细胞数明显减少(P<0.05)。结论FAM96A可抑制乳腺癌细胞增殖,诱导癌细胞凋亡,并减少侵袭转移,其低表达可能是乳腺癌发生和进展的一个机制。
Objective To analyze the expression of FAM96A in breast cancer and its effect on proliferation,apoptosis,invasion,and metastasis of breast cancer cells.Methods 75 cases of human breast cancer tissues and adjacent tissues were collected.The expression of FAM96A in breast cancer tissues and adjacent tissues was detected by Western blotting(W-B).Human breast cancer MCF-7 cells were taken as the research object and were divided into two groups,which were transfected with pcDNA3.1-vector blank plasmid and pcDNA3.1-myc-FAM96A plasmid respectively.The differences in activity,apoptosis rate,and invasion and metastasis of MCF-7 cells were compared between the two groups.Results The expression level of FAM96A protein in breast cancer tissue was(0.31±0.05),which was significantly lower than that in the adjacent tissues[(1.82±0.27)].Cell viability decreased significantly at 72 and 96 hours after overexpression of FAM96A compared with control group(all P<0.05);apoptosis rate increased after overexpression of FAM96A[(32.19±1.02)%vs.(3.07±0.84)%],and the number of perforating cells decreased significantly(P<0.05).Conclusion FAM96A can inhibit the proliferation of breast cancer cells,induce apoptosis of cancer cells,and reduce invasion and metastasis.The low expression of FAM96A may be a mechanism of the occurrence and progression of breast cancer.
