摘要
为分析西藏巨柏群体的遗传多样性,利用3个叶绿体基因(18S、CP12、CP15)针对8个群体的67份巨柏材料的叶绿体序列进行了测序分析。结果表明:3个序列的长度分别为300 bp、396 bp、1 000 bp,共检测出单倍型(H1-H11) 11个,多态性位点4个,简约性信息位点4个。表明巨柏整体遗传多样性的水平较高。同时根据AMOVA分析,发现群体内的变异也较大(85. 8%),因此针对巨柏宜采用就地保护措施。本研究对巨柏遗传资源的保护与有效利用具有重要的理论意义。
In order to research the genetic diversity of Cupressus gigantea,three chloroplast genes (18S,CP12,CP15)of 67 samples from eight populations were sequenced. The sequences length of three chloroplast genes was 300 bp,396 bp,1000 bp,respectively. A total of 11 haplotypes (H1-H11) were identified. The observed polymorphic sites and parsimony informative sites were 4 and 4,respectively. The overall genetic diversity of C. gigantea was high. The AM OVA analysis showed that the variation ratio within the population was high (85. 8%),which indicating in-situ conservation measures should be adopted for C. gigantea. These results have important theoretical significance for conservation and utilization of C. gigantea resources.
