摘要
AMP活化蛋白激酶(AMPK)是一种异三聚体的丝氨酸/苏氨酸激酶,是真核细胞中调节细胞代谢和能量稳态的关键分子。AMPK在全身组织广泛表达,低血糖、低氧、缺血和热休克等多种应激因素可将其激活,与2型糖尿病、肥胖症、恶性肿瘤等多种疾病的发生发展相关。肝脏是体内重要的代谢器官,由于其生理功能的特殊性,病原体、药物、毒物、代谢紊乱等因素均会造成肝损伤。AMPK在肝炎、非酒精性脂肪性肝病、肝纤维化、肝癌、胆汁淤积等多种急慢性肝病的发生发展中均发挥了重要作用,并有望成为部分肝病的潜在治疗靶点。
AMP-activated protein kinase(AMPK)which exists as a heterotrimer is a serine/threonine kinase and functions as the key regulator of cellular metabolism and energy homeostasis in all eukaryotic cells.AMPK is widely expressed in tissues throughout the body and can be activated by multiple stressors such as hypoglycemia,hypoxia,ischemia,and heat shock.AMPK is implicated in a number of pathological conditions,in particular type 2 diabetes,obesity and cancer.Liver is an important metabolic organ in body and may easily get harmed by pathogen,drugs,toxicant and metabolic disorders,due to the particularity of its physiological function.AMPK plays an important role in many liver diseases,such as hepatitis,non-alcoholic fatty liver disease,hepatic fibrosis,hepatocellular carcinoma,and cholestasis,and AMPK is expected to be a therapeutic target of some liver diseases.
