通督活血汤对大鼠急性脊髓损伤后炎症反应影响 被引量:3

Effect of Tongdu Huoxue Decoction on Inflammatory Response after Acute Spinal Cord Injury in Rats

摘要 目的观察通督活血汤对急性脊髓损伤后炎症因子肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)的影响,探讨通督活血汤对急性脊髓损伤后炎症反应的影响。方法 56只SPF级SD大鼠随机分为3组:假手术组(n=8)、模型组(n=24)、通督活血汤组(n=24)。模型组、通督活血汤组均采用改良Allen’s打击法建立脊髓损伤模型,假手术组不打击脊髓,其他步骤同模型组,造模后通督活血汤组给予通督活血汤灌胃,假手术组和模型组给予等量生理盐水灌胃。术后第1、3、7天对各大鼠进行BBB功能评分,评分后取脊髓组织,HE染色观察脊髓组织形态变化,Elisa法检测脊髓组织TNF-α、IL-1β含量,实时荧光定量PCR检测脊髓组织TNF-α、IL-1βmRNA的表达情况。结果与假手术组相比,术后第1、3、7天模型组、通督活血汤组BBB评分显著降低(P<0.05),与模型组相比,术后第3、7天通督活血汤组BBB评分显著增高(P<0.05);与假手术组相比,术后第1、3、7天模型组、通督活血汤组TNF-α、IL-1β及其mRNA表达量显著升高(P<0.05),与模型组相比,通督活血汤组TNF-α、IL-1β及其mRNA表达量显著降低(P<0.05)。结论通督活血汤可以降低炎症因子TNF-α、IL-1β的表达,抑制急性脊髓损伤的炎症反应,促进急性脊髓损伤后大鼠下肢功能的恢复。 Objective To observe the effects of Tongdu Huoxue Decoction on inflammatory factors tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)after acute spinal cord injury,and to explore the effect of Tongdu Huoxue Decoction on inflammatory response after acute spinal cord injury. Methods Fifty-six SPF SD rats were randomly divided into 3 groups:sham operation group(n=8),model group(n=24),Tongdu Huoxue Decoction group(n=24). The model group and Tongdu Huoxue Decoction group used the modified Allen’s method to establish the spinal cord injury model. The sham operation group did not attack the spinal cord. The other steps were the same as the model group. After the model was established,Tongdu Huoxue Decoction group was given to Tongdu Huoxue Decoction. The surgical group and the model group were given the same amount of normal saline. On the 1 st,3 rd and 7 th postoperative day,the rats were scored for BBB function. The spinal cord tissue was obtained after the score. The morphological changes of the spinal cord were observed by HE staining. The content of TNF-α and IL-1β in the spinal cord was detected by Elisa method. Real-time quantitative PCR was performed. Expression of TNF-α and IL-1β mRNA in spinal cord tissue. Results Compared with the sham operation group,the BBB scores of the 1 d,3 d,7 d model group and Tongdu Huoxue Decoction group were significantly lower(P<0.05). Compared with the model group,Tongdu Huoxue Decoction group was 3 d and 7 d after operation. The BBB score was significantly increased(P<0.05). Compared with the sham operation group,the expression of TNF-α,IL-1β and mRNA in the model group at 1 d,3 d,7 d and Tongdu Huoxue Decoction group increased significantly(P< 0.05),compared with the model group,the expression levels of TNF-α,IL-1β and mRNA in Tongdu Huoxue Decoction group were significantly lower(P<0.05). Conclusion Tongdu Huoxue Decoction can reduce the expression of inflammatory factors TNF-α and IL-1β,inhibit the inflammatory response of acute spinal cord injury,and promote the recovery of lower limb function in rats after acute spinal cord injury.
