淫羊藿苷对股骨骨折MSCs迁移及免疫功能影响 被引量:5

摘要 目的探讨淫羊藿苷对股骨骨折间充质干细胞(MSCs)迁移及免疫功能的影响。方法选取SPF级健康雄性SD大鼠96只,随机分为对照组16只及造模组80只。造模组通过截断股骨干中段建立股骨骨折模型,对照组予以假手术。将造模成功的大鼠随机分为模型组、骨肽组及淫羊藿苷低、中、高浓度组,各16只。模型组及对照组分别给予0.9%氯化钠注射液5 mL·kg^-1,日1次,腹腔注射;骨肽组给予复方骨肽注射液5 mL·kg^-1,日1次,腹腔注射;淫羊藿苷低、中、高浓度组分别给予8、16、24 mg·mL^-1的淫羊藿苷溶液5 mL·kg^-1,日1次,腹腔注射。各组大鼠均给药4周。检测比较各组骨折愈合Garrett评分、股骨骨体积(BV)、样本体积(TV)、骨体积分数(BVF)、MSCs迁移能力,以及外周血T淋巴细胞水平。结果与模型组比较,各组大鼠骨折愈合Garrett评分较高,股骨BV、TV、BVF较高,骨折端MSCs迁移能力较高,外周血CD4^+T淋巴细胞、CD4^+/CD8^+比值较高,且淫羊藿苷低浓度组<淫羊藿苷中浓度组、骨肽组<淫羊藿苷高浓度组,差异均有统计学意义(均P<0.05)。结论淫羊藿苷能有效促进股骨骨折大鼠骨折愈合,其作用可能与提高MSCs迁移能力及细胞免疫功能有关,且药物浓度越高作用越好。 Objective To analysis the effect of icariin on migration of mesenchymal stem cells(MSCs)and immune function in femoral fractur.Methods 96 SPF healthy male SD rats were selected,and were randomly divided into control group with 16 rats and model building group with 80 rats.The model building group was established femoral fractur model bycutting the middle femoral shaft,rats in the control groupreceived sham operation.The successful model of the rats were randomly divided into model group,ossotide group and icariin low,medium and high concentration group,each with 13 rats.The model group and control group were given 0.9%sodium chloride injection 5 mL·kg^-1,qd,intraperitoneal injection;ossotide group was given compound ossotide injection 5 mL·kg^-1,qd,intraperitoneal injection;icariin low-concentration group,middle-concentration group,high-concentration group were given 8,16,24 mg·mL^-1 icariin solution 5 mL·kg^-1,qd,intraperitoneal injection.All rats in each group were treated for 4 weeks.The Garrett scores of fracture healing,bone volume(BV),sample volume(TV)and bone volume fraction(BVF)of femur,MSCs migration capability,T lymphocyte levels in peripheral blood were detected and compared.Results Compared with model group,the Garrett scores of fracture healing were higher,the BV,TV and BVF of femur were higher,the migration ability of MSCs at fracture end were higher,the levels of CD4^+T lymphocyte and ratio of CD4^+/CD8^+in peripheral blood were higher in each group,and icariin low-concentration group<icariin middle-concentration group,ossotide group<icariin high-concentration group,the differences were all statistically significant(all P<0.05).Conclusion Icariin can effectively promote fracture healing in rats with femoral fracture,the effect may be related to the improvement of migration ability of MSCs and cellular immune function,and the higher the drug concentration,the better the effect.
