超声波协同酶解法对山药总多酚提取及抗氧化活性影响研究 被引量:6

Extraction and Antioxidant Activity of Total Polyphenols from Chinese Yam by Ultrasound-Assisted Enzymatic Hydrolysis

摘要 目的采用超声波协同酶解法对山药中总多酚进行测定,优化其提取工艺并对山药抗氧化活性进行初探。方法应用酶联免疫反应检测仪,采用福林酚法于760 nm波长处对山药总多酚进行含量测定,从酶的种类、酶的用量、适宜酶解pH值、酶解时间、超声提取时间等方面对山药总多酚提取工艺进行优化。以DPPH、ABTS和总还原能力为指标考察山药总多酚提取液的抗氧化能力。结果最佳提取工艺为在纤维素酶催化条件下,pH为5,酶解温度50℃,乙醇浓度65%(V/V),纤维素酶用量15 mg/g,酶解时间55 min,超声提取时间40 min时山药总多酚总提取量为10.168 mg/g,且具有良好的抗氧化能力。结论超声波协同酶解法具有提取率高、节约溶剂及时间等优点,山药中多酚类成分具有优良的抗氧化活性,为天然抗氧剂和抗氧化类保健品开发提供了新思路。 Objective To determine the total polyphenols in Chinese yam by ultrasound combined with enzymatic hydrolysis,optimize the extraction process and preliminarily explore the antioxidant activity of Chinese yam.Methods The content of total polyphenols in Chinese yam was determined at 760 nm wavelength by Folin phenol method with ELISA.The extraction process of total polyphenols from Chinese yam was optimized in terms of the type of enzyme,the amount of enzyme,the suitable pH value of enzymatic hydrolysis,the time of enzymatic hydrolysis and the time of ultrasonic extraction.The antioxidant activity of total polyphenol extract ofyam was evaluated by DPPH,ABTS and total reduction ability.Results The optimum extraction conditions were as follows:pH=5,enzymatic hydrolysis temperature 50℃,ethanol concentration 65%(V/V),cellulase dosage 15 mg/g,enzymatic hydrolysis time 55 min,ultrasonic extraction time 40 min,total polyphenols extraction amount 10.168 mg/g,and good antioxidant capacity.Conclusion Ultrasound-assisted enzymatic hydrolysis has the advantages of high extraction rate,saving solvent and time.Polyphenols in Chinese yam have excellent antioxidant activity,which provides a new idea for natural antioxidants and antioxidant health products.
