摘要
目的 观察脓毒症心肌损伤大鼠微小RNA-1(miR-1)、心型脂肪酸结合蛋白(H-FABP)、丙二醛(MDA)及Toll样受体4 mRNA(TLR4 mRNA)水平,探讨瑞舒伐他汀减轻脓毒症心肌损伤的机制.方法 30只清洁级健康雄性Wistar大鼠随机分成假手术组(Sham组)、脓毒症心肌损伤组(CLP组)、瑞舒伐他汀治疗组(ROS组),每组10只.术后24 h分别检测miR-1、H-FABP、MDA及TLR4 mRNA浓度,光镜观察心肌组织病理变化.结果 三组术后24 h H-FABP、MDA、TLR4 mRNA及miR-1水平间有显著差异(P<0.01);与Sham组同期比较,CLP及ROS组在24 h H-FABP、MDA及TLR4 mRNA水平均明显升高,miR-1水平均明显下降(P<0.01);与CLP组同期比较,ROS组H-FABP、MDA及TLR4 mRNA浓度均明显下降,miR-1水平有所升高(P<0.01).光镜下CLP组和ROS组可见心肌损伤,CLP组较ROS组损伤明显.结论 瑞舒伐他汀可影响脓毒症相关因子水平,对心肌有保护作用.
Objective The levels of microRNA-1(miR-1),Heart type fatty acid binding protein(H-FABP),MDA,Tol-like receptor 4 mRNA(TLR4 mRNA)in septic myocardial rats were observed,and to explore the mechanism of rosuvastatin(ROS)in the treatment of septic myoc ardial injury.Methods A total of thirty male Wistar rats of clean grade were randomly divided into the sham-operation group(Sham),the sepsis model group(CLP),and the Rosuvastatin treatment group(ROS),with ten rats in each group.Con-centrations of miR-1,H-FABP,MDA and TLR4 mRNA expression were detected in each group at twenty-four hours after operation,and pathological changes of myocardium were observed under light microscope.Results There were significant differences in h-FABP,MDA,TLR4 mRNA and miR-1 levels among the three groups at twenty-four hours after operation(P<0.01).Compared with the Sham group in the same period,the concentrations of H-FABP,MDA and TLR4 mRNA of CLP and ROS groups were si gnificantly increased at 24 h,while miR-1 levels were significantly decreased(P<0.01).Compared with the CLP group in the same period,the concentra-tions of H-FABP,MDA and TLR4 mRNA in the ROS group were significantly decreased,and the level of miR-1 was somewhat increased(P<0.01).Myocardial injury occured in the CLP and the ROS group under light microscope,and this injury was more severe in the CLP group than in the ROS group.Conclusion Rosuvastatin could ffect the level of sepsis-related cytokines and has a protective effect on myocardium.
