白细胞介素33对人肝癌Hep3B细胞与Huh-7细胞的增殖、迁移和侵袭的影响 被引量:2

摘要 目的探讨白细胞介素33(IL-33)对人肝癌Hep3B细胞和Huh-7细胞的增殖、迁移和侵袭能力的影响。方法取对数生长期人肝癌Hep3B细胞和Huh-7细胞,将其分为20 ng/L组、40 ng/L组、60 ng/L组及对照组。除对照组外的其他组细胞加入相应浓度的IL-33,24 h后采用四甲基偶氮唑盐法测定各组细胞的增殖能力。另取细胞,将细胞分为实验组及对照组,仅实验组加入终浓度为60 ng/L的IL-33,培养24 h后采用细胞划痕实验及侵袭实验分别检测细胞的迁移及侵袭能力。结果各浓度组的Hep3B细胞和Huh-7细胞增殖活性均高于对照组(均P<0.05),60 ng/L组的Hep3B细胞和Huh-7细胞增殖活性高于20 ng/L组、40 ng/L组(均P<0.05),但20 ng/L组与40 ng/L组差异无统计学意义(P>0.05)。与对照组比较,实验组Hep3B细胞和Huh-7细胞的迁移率和侵袭能力更低(均P<0.05)。结论IL-33可能同时具有促进人肝癌Hep3B细胞和Huh-7细胞增殖,和抑制其迁移、侵袭的作用。 Objective To investigate the effects of interleukin 33(IL-33)on the proliferation,migration and invasion capabilities of human hepatoma Hep3B cells and Huh-7 cells.Methods Human hepatoma Hep3B cells and Huh-7 cells at logarithmic growth phase were collected and were divided into 20 ng/L group,40 ng/L group,60 ng/L group and control group.Except the control group,cells in the other groups were added with corresponding concentrations of IL-33,after 24 hours methyl thiazolyl tetrazolium assay was employed to detect cell proliferation capability in each group.Another cells were collected and were divided into experimental group and control group,and only the experimental group was added with IL-33(final concentration of 60 ng/L),after 24 hours of culture wounding healing assay and Transwell assay were used to determine cell migration and invasion capabilities respectively.Results Each concentration group obtained higher proliferation activities of Hep3B cells and Huh-7 cells as compared with the control group(all P<0.05),and the proliferation activities of Hep3B cells and Huh-7 cells were higher in the 60 ng/L group than in the 20 ng/L or 40 ng/L group(all P<0.05),whereas there was no statistically significant difference between the 20 ng/L group and the 40 ng/L group(P>0.05).By comparison with the control group,the experimental group yielded lower migration rate and invasion capability of Hep3B cells and Huh-7 cells(all P<0.05).Conclusion IL-33 may promote proliferation and inhibit the migration and invasion of human hepatoma Hep3B cells and Huh-7 cells.
