摘要
目的通过生物信息学方法探讨miR-195在胃癌组织中的表达情况及其临床意义。方法下载GEO和TCGA数据库中相关数据集,比较胃癌组织和正常胃组织中miR-195表达量,并分析miR-195表达量与胃癌患者临床特征的相关性。采用生物信息学方法预测miR-195的靶基因,利用R软件的“ClusterProfiler”包对靶基因进行GO和KEGG富集分析,通过STRING 11.0在线分析网站构建PPI网络,进一步筛选出miR-195的核心基因。结果miR-195在胃癌组织中的表达水平低于正常胃组织,且对于胃癌具有较高的诊断价值(AUC>0.7,P<0.05);miR-195表达量与患者年龄、病理分级、TNM分期和淋巴结转移等密切相关(P<0.05)。GO分析显示,miR-195的靶基因主要富集在离子通道及转运蛋白、细胞发育等功能,KEGG分析主要涉及胃酸分泌、钙信号通路等。AFP、CDC6和COL1A1为PPI网络中的核心基因,在胃癌中均表达上调,其中AFP和CDC6与miR-195表达量呈负相关(P<0.05)。结论miR-195在胃癌组织中低表达,可作为抑癌基因参与胃癌的发生发展,并有潜力成为胃癌的诊断标志物及治疗靶点。
Objective To investigate the expression level of miR-195 and its clinical significance in gastric cancer tissues through bioinformatics methods.Methods The relevant data sets from GEO and TCGA databases were downloaded for the study.The expression of miR-195 in gastric cancer tissues and normal gastric tissues were then compared,and the correlation between miR-195 expression and clinical characteristics were further analyzed.The target genes of miR-195 were predicted by bioinformatics methods and analyzed by GO and KEGG analysis with the“clusterProfiler”package of R software.Furthermore,the PPI network was constructed by STRING 11.0 to identify the hub genes.Results The expression of miR-195 was decreased in gastric cancer tissues compared with normal gastric tissues,which showed a potential diagnostic efficiency for gastric cancer(AUC>0.7,P<0.05).The miR-195 expression levels were significantly associated with age,pathological grade,TNM stage,and lymph node metastasis(P<0.05).The results of GO analysis showed that the target genes of miR-195 were mainly enriched in functions such as ion channels transporters,and cell development.KEGG analysis results were mostly observed in gastric acid secretion and calcium signaling pathway.AFP,CDC6 and COL1A1 were the core genes of the PPI network and they were all up-regulated in gastric cancer.Furthermore,AFP and CDC6 were revealed to be negatively correlated with miR-195 expression(P<0.05).Conclusion The expression of miR-195 is decreased in gastric cancer tissues.It may participate in the occurrence and development of gastric cancer functioning as a tumor-suppressor,and thus it can serve as a potential diagnostic marker and therapeutic target for gastric cancer.
