生脉散对糖尿病大鼠心脏钙转运蛋白的调节作用 被引量:7

摘要 目的:探讨生脉散对糖尿病大鼠心功能和钙转运蛋白表达的影响。方法:高脂饲料喂养雄性SD大鼠4周后注射链脲佐菌素诱导糖尿病大鼠模型,将成模大鼠随机分为模型组、曲美他嗪组和生脉散组,对照组全程以普通饲料喂养,以上各组灌胃给药12周并监测大鼠体质量、血糖及动态超声心动,给药后第12周Western blot检测心肌组织钙转运相关蛋白表达水平。结果:与对照组比较,糖尿病模型大鼠第8周开始出现左心室射血分数(EF)及短轴缩短率(FS)降低,左室收缩功能降低;曲美他嗪组、生脉散组自给药第8周EF、FS增高,模型组PLB表达增高, SERCA2a、PLB Ser16+Thr17、NCX1蛋白表达降低;曲美他嗪、生脉散组给药12周后PLB Ser16+Thr17、SERCA2a、NCX1表达增加。结论:生脉散可以通过调节心脏钙转运蛋白增强肌浆网钙回摄,改善糖尿病大鼠心脏收缩功能,为临床防治糖尿病并发的心脏收缩功能损伤提供理论依据。 Objective:To investigate the effects of Shengmaisan(SMS)on cardiac contractile function and calcium transport proteins expression in diabetic rats.Methods:SD rats were fed with high-fat diet for 4 weeks,and then injected into streptozotocin to induce the diabetic model.The diabetic rats were randomly divided into Model group,Trimetazidine(TMZ)group and SMS group,besides of Control group.The Control group was fed with normal feed.Blood glucose,weight and echocardiographic were determined in this study.The expression of calcium transport protein were detected at the end of the 12th week with dynamic drug administration.Results:The left ventricular ejection fraction(EF)and short axis shortening rate(FS)of diabetic rats were significantly lower than Control group since the 8th week.EF and FS were enhanced in the SMS and TMZ group after eight weeks drug administration,compared with the Model group.The increased PLB expression and the decreased expression of SERCA2a,PLB Ser16+Thr17 and NCX1 indicate a damage of contractile function in Model diabetic heart.While SMS and TMZ can down-regulate the expression of PLB,and up-regulate the expression of PLB Ser16+Thr17,SERCA2a and NCX1.Conclusion:Shengmaipowder could improve cardiac contractile function in diabetic rats through regulating calcium transport proteins expression to enhance the sarcoplasmic reticulum calcium uptake.
