入侵我国的褐云玛瑙螺线粒体全基因系统发育分析 被引量:2

Phylogenetic analysis of the mitochondrial genomes invading giant African snail Achatina fulica in China

摘要 目的拟基于线粒体全基因组序列对我国多个地点采集的褐云玛瑙螺样本进行系统进化分析寻找该入侵物种的群体遗传结构特征。方法对采集自广西壮族自治区、广东省、贵州省、云南省、海南省与福建省共18个采样点的99份褐云玛瑙螺样本,通过二代测序获得线粒体全基因组的组装序列,并基于该序列信息利用最大似然法构建系统发育分析。结果经测序组装获得的线粒体基因组序列总长度为:15382~17125 bp,经注释包括13个蛋白编码基因序列、21个tRNA序列以及2个核糖体序列(12 S与16 S)。80份样本的线粒体全基因组与参考基因组比对呈现高度的序列相似性(100%),其余19份样本序列相似性仅有少量的差异。差异样本系统进化分析结果显示出一定的地域聚集性,但自展值较低。结论线粒体全基因组数据并不适用于入侵我国的褐云玛瑙螺群体遗传结构分析。 Objective To analyze the population genetic structure of the invasive species based on mitochondrial genome sequence analysis of the samples of Achatina fulica(A.fulica)collected from multiple locations in China.Methods A total of 99 samples of A.fulica collected from 18 sampling sites in Guangxi Zhuang Autonomous Region,Guangdong province,Guizhou province,Yunnan province,Hainan province and Fujian province were obtained.The mitochondrial genome assembly sequence was obtained by Illumina Hiseq sequencing which used to phylogenetic analysis by maximum likelihood method.Results The total length of the mitochondrial genome sequence obtained by sequencing assembly is 15382-17125 bp,including 13 protein coding gene sequences,21 tRNA sequences and two ribosome sequence(12 S and 16 S).The mitochondrial genome alignment of the 80 samples showed a high degree of sequence similarity(100%).The remaining 19 samples have only small differences in sequence similarity.The results of the evolution analysis of the differential sample showed a certain geographical clustering,but the bootstrap value was low.Conclusion The mitochondrial genome is not suitable for the genetic structure analysis of the genetic population structure of A.fulica currently invading China.
