针刺对腹型肥胖小鼠肝脏病理及糖脂代谢影响的研究 被引量:10

摘要 目的:研究针刺对腹型肥胖小鼠肝脏病理形态与糖、脂代谢的影响。方法:雄性C57BL/6J小鼠随机分为空白组6只、造模组29只,建立腹型肥胖模型,以高脂饲料喂养12周后,将造模成功的12只小鼠随机分为模型组6只和针刺组6只,继续以高脂饲料喂养。将针刺组小鼠置于固定器内固定,针刺双侧"带脉"穴,接电针,2 Hz/15 Hz疏密波,强度1.5 mA,隔日1次,每次15 min。模型组和空白组同步固定。共干预8周,HE染色观察小鼠肝脏病理形态变化;取血检测小鼠血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、游离脂肪酸(FFA)、空腹血糖值(FBG)和胰岛素(INS)含量,并计算胰岛素抵抗指数(HOMA-IR)。结果:与空白组相比,模型组小鼠血清中TC、TG、LDL-C、FBG、INS、HOMA-IR水平明显上升(P<0.01或P<0.05),血清中HDL-C含量明显下降(P<0.05);与模型组相比,针刺组小鼠血清中TC、TG、FBG、INS含量显著下降(P<0.01或P<0.05),HDL-C含量明显上升(P<0.05);血清中FFA含量呈空白组<针刺组<模型组趋势,但差异无显著性。HE染色可见模型组小鼠肝脏大量肝细胞排列紊乱,水肿和炎性细胞浸润,局部脂肪变性;针刺组肝细胞排列整齐,少量炎性细胞浸润,未见脂肪变性。结论:针刺可以调控糖、脂代谢紊乱,对肝脏病理具有良性调节作用,改善腹型肥胖。 Objective:To study the effect of acupuncture on liver pathology and glycolipid metabolism in abdominal obesity mice.Methods:Male C57BL/6J mice were randomly divided into blank group(6 mice)and model group(29 mice).Abdominal obesity models were established by feeding on high-fat diet for 12 weeks,and then the 12 mice successfully modeled were randomly divided into model group(6 mice)and acupuncture group(6 mice),and continue to be fed with high-fat diet.The acupuncture group fixed in a fixture were needled on Daimai(GB 26)on both sides,and connected with electroacupuncture,with 2 Hz/15 Hz dense-sparse wave,the intensity of 1.5 mA,once every other day for 15 min each time.The model group and the blank group were fixed synchronously.After 8 weeks of intervention,HE staining was used to observe the pathological morphology of the mouse liver;blood was taken to detect serum total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),and high density lipoprotein Cholesterol(HDL-C),free fatty acid(FFA),fasting blood glucose(FBG)and insulin(INS)content,and the insulin resistance index(HOMA-IR)was calculated.Results:Compared with the blank group,the levels of TC,TG,LDL-C,FBG,INS,and HOMA-IR in the model group increased significantly(P<0.01 or P<0.05),and the content of HDL-C in the serum decreased significantly(P<0.05);compared with the model group,the levels of TC,TG,FBG,and INS in the acupuncture group decreased significantly(P<0.01 or P<0.05),and the HDL-C content increased significantly(P<0.05);The serum FFA content showed a trend of blank grou P<acupuncture grou P<model group,but there was no significant statistical difference.HE staining showed that there were a large number of disordered liver,edema and inflammatory cell infiltration,and local steatosis cells in the model group;liver cells were neatly arranged and a small amount of inflammatory cells infiltrated,and no steatosis was seen in the acupuncture group.Conclusion:acupuncture can regulate the disorder of glucose and lipid metabolism,has a benign regulatory effect on liver pathology,and improves abdominal obesity.
