探讨双重特异性酪氨酸磷酸化调节激酶1B在宫颈癌中的潜在促癌机制

摘要 目的 探讨DRYK1B在宫颈癌中的促癌机制及其临床意义。方法 采用生物信息学数据分析及实验验证,探索DRYK1B在宫颈癌的生长、迁移、凋亡等方面的作用。结果 DYRK1B基因在宫颈鳞状细胞癌肿瘤中的表达量显著高于正常宫颈组织;DYRK1B基因高表达的患者无瘤生存率明显降低;免疫组化及Western blot实验结果显示DYRK1B在宫颈癌组织中的高表达。DYRK1B抑制剂AZ191处理宫颈癌Siha细胞系后,其生长受到抑制,迁移能力减弱,凋亡程度增加。结论 DYRK1B通过蛋白激酶相关通路促进宫颈癌细胞生长及迁移,抑制肿瘤细胞凋亡。 Objective To investigate the mechanism of dual-specific tyrosine phosphorylation-regulated kinase 1B(DRYK1B)in promoting cervical cancer and its clinical significance.Methods Biological information data analysis and experimental verification are used to explore the role of DRYK1B in growth,migration and apoptosis of cervical cancer.Results The expression of DYRK1B in cervical squamous cell carcinoma was significantly higher than that in normal cervical tissues.It may be activated by protein kinases and phosphorylation kinases to promote migration and inhibit apoptosis through protein kinase-associated signaling pathways.Patients with high expression of DYRK1B had a worse disease-free survival rate.Immunohistochemistry and Western blot analysis confirmed the high expression of DYRK1B in cervical cancer tissues.When Siha cell line was treated with DYRK1B inhibitor AZ191,its growth was inhibited,migration ability was weakened,and apoptosis was increased.Conclusion DYRK1B promotes the growth and migration of cervical cancer cells through protein kinase-related pathways and inhibits tumor cell apoptosis.
