摘要 本实验在摇瓶实验的基础上,研究了丁酸梭菌在200L罐中中试水平的发酵规律,使用RCM培养基作为种子培养基,接种量1%,200L罐装液量140L,培养温度37C,转速50rpm/min,厌氧密闭培养,在不同时间段,对单位时间补碱量、丁酸产量、活菌数、芽孢率进行研究,研究结果表明,丁酸梭菌在200L罐中发酵,延迟期5~7h,对数期10~12h,发酵20h后开始产生芽孢,最终芽孢率达到95%,自然成孢效果良好,为丁酸梭菌的规模化生产提供了一定借鉴。 In this research,on the basis of the experiment in shake flask,Clostridium butyri-cum C-101 was studied in a 200L tank on the pilot scale test.The experiment was carried out with using RCM medium as the seed culture medium,inoculated quantity 1%,140L/200L canned fluid volume,cultured temperature of 37 C,revolving speed of 50RPM/min,anaerobic culture.At different time,alkali consumption,butyric acid production,the number of living bacterium,the rate of spore was studied.The results showed that:in the 200L tank,the Clostridium butyricum,lag phase maintained 5~7 h,logarithmic phase of 10~12 h,fermented after 16h,spores come into existence,the spore rate eventually reach of 95%.The natural sporulation effect acted well,which provides reference for the large-scale production of Clostridium butyrate.
