摘要
目的探讨微小RNA-200a(miRNA-200a)对淋巴瘤DOHH-2细胞增殖的影响及其机制。方法在淋巴瘤细胞DOHH-2中转染miRNA-200a,采用qRT-PCR和Western印迹的方法检测DOHH-2细胞进行转染的24 h后的miRNA-200a和PTEN mRNA和蛋白的表达情况,采用CCK8检测各组细胞增殖,采用划痕实验检测各组细胞迁移细胞速率,每组重复6次。结果转染miRNA-200a后,与对照组或阴性对照组比,miRNA-200a转染组的miRNA-200a水平显著增高(P<0.05),PTEN水平显著降低(P<0.05),对照组和阴性对照组的miRNA-200a及PTEN mRNA及蛋白表达差异无统计学意义(P>0.05),转染miRNA-200a后,与对照组或阴性对照组比,miRNA-200a转染组的相对细胞活力和细胞迁移速率明显增高(P<0.05)。结论miRNA-200a调控淋巴瘤细胞DOHH-2的增殖与迁移,其作用机制与miRNA-200a反向调控肿瘤抑制因子PTEN有关。
Objective To investigate the effect of microRNA-200 a on the proliferation and migration of lymphoma DOHH-2 cells and its mechanism.Methods The miRNA200 a was transfected into lymphoma cell line DOHH-2.The mRNA and protein expression of miRNA200 a and PTEN were detected by Western blot and qRT-PCR in lymphoma DOHH-2 cells after 24 h of transfection.The CCK8 was used to detect each group.The cells were proliferated,and the cell migration rate of each group was measured by a scratch test,and each group was repeated 6 times.Results Compared with the control group or the negative control group,the miRNA200 a level in the miRNA200 a transfection group was significantly increased(P<0.05),PTEN level was significantly decreased(P<0.05).The miRNA200 a and PTEN mRNA and protein in the control group and the negative control group were comparable with the control group or the negative control group(P>0.05).After transfection of miRNA200 a,the relative cell viability and cell migration rate of miRNA200 a transfection group were significantly higher than those of the control group or negative control group(P<0.05).Conclusions MicroRNA-200 a regulates the proliferation and migration of DOHH-2 lymphoma cells,and its mechanism is related to the reverse regulation of PTEN by microRNA-200 a.
