南蛇藤总萜类化合物对人脐静脉内皮细胞增殖抑制及诱导凋亡的实验研究

Effect of Dulcamara Terpenoid Inhibit Proliferation and Induce Apoptosis on HUVECs

目的:研究南蛇藤总萜类化合物对人脐静脉内皮细胞增殖和凋亡的影响。方法:南蛇藤总萜类化合物作用人脐静脉内皮细胞后,MTT法检测对细胞增殖的影响;流式细胞仪法检测细胞凋亡以及基因蛋白Bcl-2表达的变化;ELISA法检测VEGF分泌含量的变化。结果:20、40、80、160μg/mL不同浓度的南蛇藤总萜类化合物作用人脐静脉内皮细胞24 h后,其抑制率分别为6.11%、13.59%、22.48%、29.61%,呈剂量依赖性。南蛇藤总萜类化合物可以诱导人脐静脉内皮细胞凋亡,随着药物浓度不断增大,其凋亡率也在逐步升高,并且存在时间依赖性。能明显下调基因蛋白Bcl-2和血管内皮生长因子(VEGF)的表达,且与药物的浓度呈负相关。结论:南蛇藤总萜类化合物能够抑制人脐静脉内皮细胞增殖,诱导其细胞凋亡,并存在一定的量效关系。其机制可能与下调Bcl-2和VEGF的表达有关。

Objective To evaluate the effect of dulcamara terpenoid on proliferation and cell apoptosis of HUVECs.Methods MTT assays were performed to study the toxic action of dulcamara terpenoid on HUVECs,and flow cytomete assay was used to test cell apoptosis effect of dulcamara terpenoid on HUVECs.And flow cytomete assay was used to evaluate the expression change of gene proteinum Bcl-2 and ELISA assay was used to test the level of VEGF.Results Dulcamara terpenoid inhibited HUVECs proliferation and cell apoptosis was to be encouraged,when treated with the dulcamara terpenoid with the range of certain concentration(20、40、80、160μg/mL),the suppression rate was 6.11%、13.59%、22.48%、29.61%,respectively,was dose-dependent.Dulcamara terpenoid can down regulated the expression of Bcl-2 and VEGF with a dose manner.Conclusion Dulcamara terpenoid can inhibit the proliferation and induce the apoptosis of HUVECs.The mechanism may be related to the down-regulation of bcl-2 and VEGF expression.
