摘要
多层滴丸是可以使双歧杆菌活菌到肠的制剂技术,但现有的检测手段尤其是样品前处理的方法无法对其含有的活菌数进行有效检测。为此,我们优化了长双歧杆菌R175活菌数检测方法,使其可以有效检测。采用了基于中心复合设计的响应面法优化了影响样品前处理结果的因素包括均质速度、均质时间、水溶时间。优化后的检测方法如下:使用Mitsuoka’s buffer缓冲液,对样品进行水浴和均质处理,具体参数为37℃水浴11 min 44 s,12462 r/min均质5 min 17 s,并采用TOSP培养基。利用响应面法成功地优化了多层滴丸中长双歧杆菌R175活菌数检测方法,可以有效地用于多层滴丸中长双歧杆菌R175的活菌计数。
Multilayer dripping pills as the preparation technology could bring live Bifidobacteria into the intestine.However,the existing detection methods,especially sample pretreatment methods,could not detect the number of viable bacteria effectively.In order to detect the viable number of Bifidobacterium longum R175 effectively,the detection method for live bacteria counting in multilayer dripping pills were optimized.Response surface methodology based on central composite design was used for the optimization of factors affecting sample pretreatment,such as homogenization speed,homogenization time and water solubility time.The optimized results were as follows:samples treated with dissolution buffer,water bath at 37℃for 11 min 44 s,then homogenized with 12462 r/min for 5 min 17 s,and used with TOSP medium.As a result,the detection method for counting the number of viable Bifidobacterium longum R175 in multilayer dripping pills was successfully optimized by response surface method,which could be effectively used for counting the number of viable Bifidobacterium longum R175 in multilayer dripping pills.
